Gene expression profiles of neonatal cardiomyocytes and immortalized heart muscle cells exposed to inflammatory stimuli

ABSTRACT: We used a transcriptomic approach to systematically compare the response to inflammatory stimuli for two murine cardiomyocyte models. Our results indicate that murine neonatal cardiomyocytes (NNC) are able to exhibit a pronounced response upon inflammatory stimuli, while HL-1 cells were predominantly recruiting transcripts corresponding to Jak-Stat-signalling.

Specific Inflammatory Stimuli Lead to Distinct Platelet Responses in mice and Humans

Project description:Different inflammatory stimuli contribute to the formation of atherosclerosis. It is hypothesized that although the end result is the same - plaque formation in arterial vessels - the pathogenesis is dependent on the etiology. In particular, platelets will respond differently depending on the inflammatory stimuli and timepoint. Using a microarray and platelet inflammatory function studies, we identified the transcriptional and functional changes that occur early and late with different inflammatory stimuli. ApoE-/- C57BL/6 mice were left untreated (control) or administered oral P. gingivalis (Pg) or intranasal C. pneumoniae (Cp) for 3 weeks, and sacrificed either 1 day (early timepoint) or 9 weeks (late timepoint) after this 3-week period. A separate group of animals was fed a Western Diet (WD) for 9 weeks and then sacrificed. Whole blood samples were collected from each animal into citrate solution and serially centrifuged to produce a pure platelet population. RNA was extracted and pooled from each experimental group and hybridized to Affymetrix Mouse Gene 1.0 ST microarrays.

Metabolomic profiling of macrophages determines the discrete metabolomic signature and metabolomic interactome triggered by polarising immune stimuli

Project description:Priming and activating immune stimuli have profound effects on macrophages, however, studies generally evaluate stimuli in isolation rather than in combination. In this study we have investigated the effects of pro-inflammatory and anti-inflammatory stimuli either alone or in combination on macrophage metabolism. These stimuli include host factors such as IFNγ and ovalbumin-immunoglobulin immune complexes, or pathogen factors such as LPS. Untargeted LC-MS based metabolomics provided an in-depth profile of the macrophage metabolome, and revealed specific changes in metabolite abundance upon either individual stimuli or combined stimuli. Here, by factoring in an interaction term in the linear model, we define the metabolome interactome. This approach allowed us to determine whether stimuli interact in a synergistic or antagonistic manner. In conclusion this study demonstrates a robust approach to interrogate immune-metabolism, especially systems that model host-pathogen interactions.

Gene expression response to 5 microbial stimuli in Detroit562 cells [RNA-seq]

Project description:NFKB is a family of transcription factors (TF), which are master regulators of the innate immune system. It is activated downstream of pathogen recognition receptors after ligand binding and regulates the expression of antimicrobials, cytokines and chemokines, thus helping to fight infections as well as recruiting the adaptive immune system. Although NFKB responds to a wide variety of signals, the processes in which stimulus-specificity is attained are still unclear. We characterized the response of one of the NFKB members, RELA, to five stimuli mimicking infection in nasopharyngeal epithelial cells. We wanted to investigate gene expression in response to these stimuli as well, in order to relate RELA binding to differences to expression regulation. Although most RELA binding sites where common among stimulation, we distinguished a minority of stimulus-specific RELA binding sites. Interestingly, some of these seemed to have a biological effect as they correlated with corresponding gene expression. Especially, the response to Poly I:C mimicking viral dsRNA gave a distinct RELA profile binding the vicinity of antiviral genes. Some of which were overexpressed under Poly I:C stimulation specifically.

Binding Profiles of Myc and RNA Polymerase II in Rat Neonatal Cardiomyocytes

Project description:During the progression of cardiac hypertrophy, cardiomyocytes accelerate RNA biogenesis to meet increasing metabolic demands under exogenous hypertrophic stimuli. We have elucidated the quantitative kinetics of RNA in c-Myc (Myc)-mediated hypermetabolic analytical model using cultured neonatal rat cardiomyocytes. Chromatin immunoprecipitation and high-throughput sequencing demonstrate that overexpressed Myc binds to a specific set of genes directly in cardiomyocytes and recruits RNA polymerase II to these genes.
