Project description:Anaylsis differentially expressed genes of mouse peri-implanted uteri comparing pre-implantation uteri (Day2, Day3 and Day4) with post-implantation uteri (Day6, Day7 and Day8) by microassay. This study has built a meaningful basis for future investigation in elucidating the molecular nature of maternal-fetal interactions during pregnancy establishment and maintenance. Pre-implantation uteri VS. Post-implantation uteri. Three biological replicates of each experiments: pre-implantation uteri (Day2, Day3 and Day4): 234, 234①, 234②; 3 mixture of post-implantation uteri (Day6, Day7 and Day8): 678, 678①, 678②. Two hybridizations were performed by using a reverse fluorescence strategy (Cy3, Cy5) for each sample.

Coordinated metabolic transitions and gene expression by NAD+ during adipogenesis.

Project description:Adipocytes are the main cell type in adipose tissue, a critical regulator of metabolism, highly specialized in storing energy as fat. Adipocytes differentiate from multipotent mesenchymal stromal cells through adipogenesis. Using RNA-seq we explored the effect of manipulating NAD+ bioavailability during adipogenic differentiation from human mesenchymal stem cells. Cells were harvested either undifferentiated (hMSC) or after days 8 and 16 of adipogenic induction. During adipogenesis, cells were treated with either NAD+ (5 mM), or FK866 (1 nM) or EX527 (50 μM).

Single-cell profiling of human adipose tissue-derived mesenchymal progenitor cells induced toward adipogenic fate.

Project description:To understand the specific transcriptomic changes in cells undergoing adipogenic differentiation, we performed single cell RNA-seq on two separate cultures: one corresponding to multipotent progenitors grown to confluency but not subjected to differentiation stimuli, and the second corresponding to progenitors exposed to adipogenic media for 3 days. Homogenous multipotent progenitors underwent two distinct fate trajectories rapidly upon induction of adipose differentiation, one toward the adipocyte fate, and the other towards a distinct, undifferentiated state.
