Project description:To evaluate involvement of miR-221 and miR-222 in lung cancer, we investigated the effects of miR-221 and miR-222 overexpression on six lung cancer cell lines as well as one immortalized normal human bronchial epithelial cell line. Two cell lines, H3255 and H1299 with no replicates were studied. Cells were transfected with miR-221, miR-222, or miR control. Microarray analysis was done to identify genes differentially expressed in lung cancer cells after the transfection of miR-221 or miR-222.

Project description:MicroRNAs (miRNAs) constitute fine tuners of gene expression and are implicated in a variety of diseases spanning from inflammation to cancer. miRNA expression is deregulated in rheumatoid arthritis (RA), however, their specific role in key arthritogenic cells such as the synovial fibroblast (SF) remains elusive. We have shown in the past that the expression of the miR-221/222 cluster is upregulated in RA SFs. Here, we demonstrate that miR-221/222 activation is downstream of major inflammatory cytokines, such as TNF and IL-1β, which promote miR-221/222 expression independently. miR-221/222 expression in SFs from the huTNFtg mouse model of arthritis correlates with disease progression. Targeted transgenic overexpression of miR-221/222 in SFs of the huTNFtg mouse model led to further expansion of synovial fibroblasts and disease exacerbation. miR-221/222 overexpression altered the transcriptional profile of SFs igniting pathways involved in cell cycle progression and ECM regulation. Validated targets of miR-221/222 included p27 and p57 cell cycle inhibitors, as well as Smarca1 (a chromatin remodeling component). In contrast, complete genetic ablation of miR-221/222 in arthritic mice led to decreased proliferation of fibroblasts, reduced synovial expansion and attenuated disease. scATAC-seq data analysis revealed increased miR-221/222 gene activity in the pathogenic and activated clusters of the intermediate and lining compartment. Taken together, our results establish an SF-specific pathogenic role of the miR-221/222 cluster in arthritis and suggest that its therapeutic targeting in specific subpopulations should inform the design of novel fibroblast-targeted therapies for human disease.

Project description:MicroRNAs (miRNAs) regulate the expression of mRNAs through sequence-specific binding into their 3′ untranslated region (UTR). The seed sequence of miRNAs is the key determinant to recognize the target sites. The paralogous miRNAs, which share the same seed sequences but differ in their 3′ parts, are known to regulate largely overlapping group of miRNAs. However, there is still no study which analyzes the functional difference among paralogous miRNAs. In this study, we compared the function between paralogous miRNAs, miR-221 and miR-222. By employing nuclease-mediated genome engineering technique, we established the knockout cell lines for these miRNAs, and analyzed their difference in target regulation precisely. We found that miR-221 and miR-222 suppress the previously identified targets, CDKN1B and CDKN1C, differentially. From the transcriptome analyses, we also found that large number of different transcripts with independent functions respond exclusively only to each of miR-221 and miR-222, respectively. Therefore, the miRNAs with common seed sequences can exert dissimilar function by regulating different groups of target mRNAs. This study illustrates that more researches are required to establish the rules of target site recognition by miRNAs.

However, the differentiation pattern and underlying mechanisms remain unclear. Here, we find that cAMP/PKA/CREB1-induced miR-221/222 suppression contributes to the neuron-like differentiation of gliomas. cAMP agonists selectively induced neuron- and astrocyte-like but not oligodendrocyte-like differentiation of C6 glioma cells. PKA inhibitors and CREB1 knockout blocked neuron-like differentiation of glioma cells. cAMP inhibited miR-221/222 in a PKA/CREB1 dependent manner. Importantly, both in vitro and in vivo assays demonstrated that transcriptional suppression of miR-221/222 is required for neuronal differentiation of glioma cells. Our findings suggest that increasing cAMP levels can induce bidirectional differentiation of glioma cells. Furthermore, the miR-221/222 cluster acts as an epigenetic brake during glioma differentiation. Project description:Human bone marrow mesenchymal stromal cells are capable of limited self-renewal and multi-lineage differentiation in vitro. Several studies have demonstrated that microRNAs, post-transcriptional modifiers of protein expression, play crucial roles in the regulation of these complex processes. To gain knowledge regarding the role of microRNAs in human adipocyte regulation, we examined the microRNA expression profile of the immortalized human bone marrow-derived stromal cell line hMSC-Tert20. We identified 12 microRNAs that were differentially expressed during adipogenesis, of which several have previously been shown to play important roles in adipocyte biology. The expression of miR-155, miR-221 and miR-222 decreased during the adipogenic program, suggesting that they act as negative regulators of differentiation. Interestingly, adenovirus-mediated expression of either miR-155 alone or miR-221 plus miR-222 significantly inhibited adipogenesis and repressed induction of the master regulators C/EBP? and PPAR?. Our study provides the first experimental evidence that miR-155, miR-221 and miR-222 function in human adipocyte differentiation. The telomerase immortalized human bone marrow-derived stromal cell line hMSC-Tert20 was differentiated towards adipocytes, and total RNA was harvested at various time points (0, 8, 24, 32, 48 and 72 hours, and 7, 14 and 21 days). All hybridizations, except for 0h, 8h and 32h, were performed twice. The expression of each miRNA at a given time point was calculated as a ratio relative to its level on day 0. No dye swaps.
