Project description:T2 progenies of two transgenic lines overexpressing ERF transcription factor WIN1 were grown on soil in parallel under identical conditions. mRNA was extracted from pooled leaves from multiple plants of each line for the microarray experiement. Plants derived from two lines were used in this experiment: line 13, a "medium" overexpressor, and line 22, a strong overexpressor. Control mRNA was obtained from pooled control plants transformed using an empty binary vector. All plants were first selected on medium containing kanamycin before being transferred to soil. Project description:Aerial parts of the rice-Arabidopsis FOX (Full-length cDNA overexpressor) lines K16331 and K19624 harboring the rice FL cDNA of LBD37 (Os-LBD37) were analyzed. LBD37 belongs to the plant- specific LOB- (Lateral Organ Boundary) domain family proteins first characterized in Arabidopsis. Results point towards an involvement of the rice LBD37 ortholog of Arabidopsis in nitrogen metabolism- related processes. K16331 (T2) and empty vector control plants (T2) were grown for 18 days on solid MS- medium supplemented with 20mg/l Hygromycin. For each hybridization on Affymetrix microarrays, 2 independent plants for K16331 were used. Homozygote K19624 (T3) and empty vector control plants (T3) were grown for 18 days on solid MS- medium. For each hybridization on Affymetrix microarrays, 1 plant was used.

Project description:In higher plants, various developmental and environmental conditions enhance expression of the mitochondrial alternative oxidase (AOX). In this work transgenic Arabidopsis thaliana plants were generated that either overexpress AOX or inhibit its synthesis. Gene expression following antimycin A treatment, which inhibits the cytochrome pathway in mitochondria, was studied in an AOX overexpressor line. The role of AOX in regulation of reactive oxygen species (ROS) in leaves was studied in the transgenic lines. The transgenic lines were also used to investigate the mitochondria-chloroplasts interaction in assays performed at three-times growth light. For most of the parameters measured AOX antisense lines and WT plants showed a very similar response whereas AOX sense lines differed in several aspects. An unexpected result was that AOX overexpressors had higher cellular ATP/ADP ratios as compared to WT and antisense lines. This was linked to decreased photosynthetic CO2-assimilation and O2-evolution, closed PSII reaction centers and altered Calvin cycle metabolite contents. We conclude that increased mitochondrial AOX capacity can lead to a drain of electrons from the chloroplasts causing an unfavorable redox status and thereby inhibit photosynthesis. Project description:Transcriptional profiling of Arabidopsis thaliana 12-days old seedlings comparing Col-0 wild type with transgenic plants with altered expression of dual-targetting plastid/mitochondrial organellar RNA-polymerase RPOTmp. Transgenic plants used for experiment were: overexpressor plants obtained by transformation of Col-0 WT plants with genetic constructs created in [Tarasenko et al., 2016] contained catalytic part of RPOTmp enzyme with transit peptides of RPOTm (mitochondrial) and RPOTp (plastid) by agrobacterial transformation; plants with complementation of RPOTmp functions in mitochondria or chloroplasts obtained from transformation of GABI_286E07 rpotmp knockout-mutant plants with genetic constructs created in [Tarasenko et al., 2016]. Goal was to determine the effects of RPOTmp knockout/overexpression on global Arabidopsis thaliana gene expression.

Hygromycin- resistant rice Os-LBD37 overexpressor and empty vector control plants were grown in hydroponic culture system. The sampling date of the leaf blades of RK16331-13 LBD37 overexpressor plants and the empty vector control was determined according to the emergence of the 8th leaf blade. Three independent hybridizations were performed for line RK16331-13 and the empty vector control line.

Project description:Our previous experiment on biogenesis mechanisim of stress-inducible antisense RNAs showed that accumulation of antisense RNA in a drought-inducible gene, RD29A locus was decreased in rdr1/2/6 triple mutant. The RD29A antisense RNA did not have CAP structure and poly(A) tail. We applied deep sequencing of uncapped RNA using SOLiD to identify RDR1/2/6-mediated antisense RNAs in genome-wide. We identified 1) 975 RDR1/2/6-mediated antisense RNA loci on which no. of sequence tags is decreased in rdr1/2/6 and makes peak and 2) 1,613 RDR1/2/6-mediated antisense RNA loci on which no. of the tags is decreased in rdr1/2/6 and enriched on antisense strand of exons. They were grouped into 1,959 non-redundant loci.
