MicroRNA profiling in human colon cancer cell lines

Project description:MicroRNA profiling in colon cancer cell lines. Two cell lines have been found with impairment on the export of precursor miRNAs into the cytoplasm (XPO5), HCT15 and DLD1. To test the microRNAs involved several comparisons have been performed: DLD1 and HCT15 cell lines have been compared with XPO5 wild-type cell lines (HCT116 and RKO). Furthermore the transfected XPO5 wild-type defective cells (DLD1.XPO5 wt and HCT15.XPO5 wt) have been compared respect to the non-transfected cells (HCT15 and DLD1). Finally the cell lines HCT15 and DLD1 transfected with a negative control (HCT15.XPO5 358-366 and DLD1.XPO5 358-366) have been compared with respect to non-transfected cell lines (HCT15 and DLD1).

Transcriptomic analysis of trametinib-resistant HCT116 colorectal carcinoma cells compared to the parental control cells

Project description:HCT116 cells were treated with with increasing concentrations of trametinib over 2 months. Drug-resistant clones emerged and were cultured in the presence of 30 nmol/L trametinib. These cells exhibited a greater than 10-fold increase in the GI50 for trametinib compared to the parental cell line. RNA-seq of the resistant clone HCT116_R4 versus the parental cells identified differentially expressed genes potentially involved in resistance.

Role of miR-19b and its target mRNAs on 5-fluorouracil resistance in colon cancer cells

Project description:Despite the efforts in defining the molecular mechanisms for the drug resistance in colorectal cancers, little is known about the roles of microRNAs. With microarray containing 723 microRNAs, we examined effect of 5-fluorouracil (5-FU) on the microRNA expression. Respond to 5-FU, we identify two microRNAs, miR-19b and miR-21, that were differentially expressed in 5-FU resistant colon cancer cells derived from KM12C and DLD-1. DLD-1, DLD-1/R, KM12C, and KM12C/R cells were plated at 1 × 105 cells/well. After pre-culture, cells were treated with 60 uM of 5-FU for 72 h. This was the same condition as the analysis of cell cycle. RNAs were collected before (0 h) and after the treatment of 5-FU (72 h).
