Transcriptomic signatures in whole blood of patients who suffer a Chronic Inflammatory Response Syndrome (CIRS) as a result of exposure to the marine toxin ciguatoxin

ABSTRACT: This study compares the transcriptional profiles of 11 patients with ciguatera induced Chronic Inflammatory Response Syndrome to 11 normal controls using whole blood perserved in PAXgene blood collection tubes and Agilent whole genome microarrays. In our analysis, we found nearly 200 genes significantly, differentially expressed in patients compared to controls.

Genome-wide analysis of the effect of long-term freezing of PAXgene Blood RNA tubes

Project description:Analysis of long-term freezing on the stability of transcriptome profiles in PAXgene stabilized whole blood samples. In the present study it was tested if long-term freezing of PAXgene RNA tubes (up to one year) has an influence on the transcriptome profile of peripheral whole blood samples. Results indicated that gene expression profiles of whole blood samples stabilized with PAXgene RNA tubes remain stable for at least 1 year. Total RNA was obtained from peripheral whole blood samples collected in PAXgene RNA tubes of healthy human subjects. RNA was either isolated after 24h storage at room temperature or after freezing of PAXgene RNA tubes for up to 1 year (6 weeks, 6 months and 12 months).

Gene expression analysis of peripheral whole blood RNA following ischemic stroke

Project description:The purpose of this project was to elucidate gene expression in the peripheral whole blood of acute ischemic stroke patients to identify a panel of genes for the diagnosis of acute ischemic stroke. Peripheral blood samples were collected in Paxgene Blood RNA tubes from stroke patients who were >18 years of age with MRI diagnosed ischemic stroke and controls who were non-stroke neurologically healthy. The results suggest a panel of genes can be used to diagnose ischemic stroke, and provide information about the biological pathways involved in the response to acute ischemic stroke in humans.
