MicroRNA expression profile of PBMCs in OAD mice and control mice

ABSTRACT: MicroRNA expression profiling was performed in PBMCs collected from 12 C57B/6 mice at 7 and 15 days after administration of H2Kb and Isotype antibody. Results: there was a substantial proportion of microRNAs changed over time in OAD mice compared to control mice. MHC Class I antibody induced gene expression in mouse obliterative airway disease

Project description:To further understand the early molecular events in antibody mediated chronic inflammation of the lungs, we utilized a murine Obliterative Airway Disease (OAD) model of native lungs where Abs to MHC class I induces bronchiolar obliteration and fibrosis. This study identified a unique transcriptional profile with a set of genes that were stimulated while another set of genes that were repressed in anti MHC I treated lungs compared to that of isotype treated lungs. Lung transplantation (LTx) is an accepted clinical intervention for various intractable pulmonary dysfunctions. However, de novo Abs to mismatched donor HLA (DSA) predispose lung-grafts to chronic rejection, Bronchiolitis Obliterans Syndrome (BOS). The anti-MHC induced mouse OAD model, the only available preclinical model for study of early pathogenesis following MHC ligation, produces lesions analogous to BOS.

DNA Methylation Profiles of Airway Epithelial Cells and PBMCs from Healthy, Atopic and Asthmatic Children.

Project description:DNA methylation profiling of airway epithelial cells (AECs) and peripheral blood mononuclear cells (PBMCs) from normal, atopic and asthmatic subjects. The Illumina GoldenGate Methylation Cancer Panel I was used to obtain DNA methylation profiles across approximately 1505 CpGs in AECs and PBMCs. Samples included 7 healthy, 9 atopic, 4 atopic asthmatic and 5 non-atopic asthmatic subjects. Please note that only some of the samples are matched (i.e. AECs and PBMCs from the same individual) due to DNA quality or sample collection (i.e. only one sample (AEC or PBMC) was collected from the patient).

Project description:Social stress mouse models were used to simulate human post-traumatic stress disorder (PTSD). C57B/6 mice exposed to SJL aggressor mice exhibited behaviors accepted as PTSD-in-mouse phenotype: 'frozen' motion, aggressor's barrier avoidance, startled jumping, and retarded locomotion. Transcripts in spleen, blood and hemi-brain of stressed and control C57B/6 mice were analyzed using Agilent's mouse genome-wide arrays.

Effects of blocking IL-17A on lung gene expression after ozone exposure in obese (db/db) mice

Project description:Obese mice have an augmented response to Ozone exposure (2 ppm for 3 hours and studied 24 hours later), including increase airway hyperresponsiveness and BAL inflammation when compared to WT ozone exposed mice. When IL-17A is block with systemic injected of IL-17A antibodies there is a decrease of response in the obese mice. To determine the mechanism of action, RNA was extracted from the lungs of IL-17A and Isotype injected ozone exposed obese mice and microarray analysis of gene expression was performed.
