3' RNA-seq of fly expressing cabut RNAi after exposure to different levels of sucrose

ABSTRACT: Control (+/cbtE1-UAS-cbt RNAi) or cabut RNAi flies (Tim-gal4, UAS-cbt RNAi) were starved for 16 hours and then exposed to food containing different concentrations of sucrose: 0, 25, 50 and 100 % for 18 hours. Fly heads were collected, RNA was extracted and RNA-seq libraries were prepared as previously described (Engreitz et al., 2013)

Expression data from Drosophila mutantsÂ´ (misexpression and overexpression) of cabut/dTIEG gene

Project description:cabut (cbt), is the Drosophila ortholog of the human TGF-Î² Inducible Early Gene 1 (dTIEG). It encodes a transcription factor involved in Drosophila dorsal closure (DC), and it is expressed in embryonic epithelial sheets and yolk cell during this process upon activation of the Jun N-terminal kinase (JNK) signaling pathway. Additional studies suggest that cbt may have a role in multiple developmental processes. We used microarrays to detail the target genes of Cbt during dorsal closure process cbtEP(2)2237E1 homozigote Drosophila mutant embryos were selected at 10-13 stage (during dorsal closure process) from non-GFP embryos from the line cbtEP(2)2237E1/Cy0, twi-eGFP line by embryo sorter assays. In parallel, samples of white embryos at same stages (as control) were sorting also by embryo sorter. Also using UAS-GAL4 system, Cbt was overexpressed in the embrionic epidermis during 10-13 stage by epidermal driver (69B-Gal4) and UAS-Cbt line.

Project description:Samples 1-8: Tissue-specific RNA sequencing (Illumina) using dissected ring glands isolated from TWO different time points of control (phm>w1118) third instar larvae. Time points are: light phase zt0-4 (which corresponde to 2-4 hours from second to third instar larvae molt); and dark phase zt18-22 (which corresponde to 16-20 hours from second to third instar larvae molt) Samples 9-32: Tissue-specific gene expression (RNA seq Illumina) using dissected ring glands isolated from TWO different time points of third instar larvae. Genotypes were Timeless-RNAi (phm>tim-RNAi), Period-RNAi (phm>per-RNAi), UAS-TimcDNA (phm>UAS-Tim) and UAS-TimcDNA;UAS-PercDNA (phm>UAS-TimcDNA;UAS-PercDNA). Goal was to identify circadin pathway dependent gene sets in the ring gland. Time points were 2-4 hours and 18-20 hours after L2-L3 molt.

Project description:List of protein scores for putative TANGO10 interactors from mass spectrometry analysis of FLAG-tagged TANGO10 using either elav-GAL4 or tim-GAL4 at both ZT10 and ZT 22, as determined using Proteome Discoverer software (ThermoFisher). Fly heads from GAL4 heterozygotes were used as controls. The hits from FLAG-tagged twenty-four (TYF) proteomics were also excluded from the list to increase TANGO10-specificity. Proteins listed are those present in at least one elav-GAL4 sample and at least one tim-GAL4 sample but no negative controls.
