Changes of genes in time-dependent photoaged mouse model

ABSTRACT: Photoaging results from the damaging effects of long-term exposure to UV. It is characterized by deep wrinkle, but the mechanism is still lack. To better understand molecular events contributing to photoaging, We used microarray analysis using an optimized in time-dependent wrinkle model in mice.

Effect of rice fermentation and succinic acid on gene expression of human dermal fibroblasts irradiated with UVA

Project description:Skin aging caused by UV is called photoaging, which is characterized by deeper wrinkles accompanied by senescence of dermal fibroblasts and reduction of collagens. Rice fermentation, widely used in the cosmetics, has been reported to possess anti-aging benefits on skincare, however, its roles in the skin photoaging remains unclear. In this study, the effects of Maifuyin (a rice fermentation), and its ingredients succinic acid (SA) and choline on UVA-induced senescence in fibroblasts were evaluated. A mRNA sequencing technology (RNA-seq) was applied to study the effects of these ingredients on UVA-induced photoaging. In conclusion, these results highlight the potential use of Maifuyin and SA as promising agents for anti-photoaging applications.

Project description:Skin damage from solar ultraviolet radiation (UVR) accumulates in the dermal extracellular matrix (ECM) and contributes to photoaging. Following UVR exposure, matrix metalloproteinases (MMPs) are secreted by dermal fibroblasts to repair and remodel the ECM. Molecular signaling pathways delineating the induction of MMPs are currently well-defined; however, the effects of UV exposure on epigenetic mechanisms of MMP induction are not as well understood. An epigenetic mechanism would further describe how MMP genes are regulated in response to UV. In this study, we examined solar simulated UVR (ssUVR)-induced gene expression changes and alterations to histone methylation in the promoters of MMP1 and MMP3 in primary human dermal fibroblasts (HDF). This set of gene expression data was generated to identify photoaging related genes (including MMP) that were impacted by ssUVR exposure in our system.

Single-cell RNA-seq of UVB-radiated Skin Reveals Landscape of Photoaging-related Inflammation and Protection by Vitamin D

Project description:Due to the destruction of the ozone layer, the ultraviolet radiation received by our skin has increased significantly. Continuous exposure to sunlight can cause damage to our skin, which can lead to photoaging and even skin cancer. We performed single-cell RNA sequencing on the skin of normal and UV-irradiated mice, and found that UV-irradiation caused an inflammatory response in the skin of mice. On the contrary, the inflammation of the mice injected with vitamin D was significantly reduced. Finally, we uncovered that Hmox1 and vitamin D is related to achieve the anti-inflammatory effect of vitamin D. This provides a strategy for skin anti-UV treatment

Alterations in Extracellular Matrix Composition during Aging and Photoaging of the Skin

Project description:Human skin is composed of the cell-rich epidermis, the extracellular matrix (ECM) rich dermis, and the hypodermis. Within the dermis, a dense network of ECM proteins provides structural support to the skin and regulates a wide variety of signaling pathways which govern cell proliferation and other critical processes. Both intrinsic aging, which occurs steadily over time, and extrinsic aging (photoaging), which occurs as a result of external insults such as UV radiation, cause alterations to the dermal ECM. In this study, we utilized both quantitative and global proteomics, alongside single harmonic generation (SHG) and two-photon autofluorescence (TPAF) imaging, to assess changes in dermal composition during intrinsic and extrinsic aging. We find that both intrinsic and extrinsic aging result in significant decreases in structural ECM integrity, evidenced by decreasing collagen abundance and increasing fibril fragmentation, and ECM-supporting proteoglycans. Intrinsic aging also produces changes distinct from those produced by photoaging, including reductions in elastic fiber and crosslinking enzyme abundance. In contrast, photoaging is primarily defined by increases in elastic fiber-associated protein and pro-inflammatory proteases. Changes induced by photoaging are evident even in comparisons of young underarm and forearm skin, indicating that photoexposure experienced by an individual’s mid-20s is sufficient for large-scale proteomic alterations and that molecular-level changes due to photoaging are evident well before clinical indications are present. GO term enrichment revealed that both intrinsic aging and photoaging share common features of chronic inflammation.

Analysis of time-resolved immunological phases in the NIF mouse model for liver fibrosis.

Project description:A time-course, bulk RNA-sequencing analysis of liver and kidney tissue from NIF mice was performed to identify the dynamics of key processes contributing to the pathogenesis. Transcriptomics profiling of the NIF mouse revealed a gradual transition from an initially reactive inflammatory response to a regenerative, pro-fibrotic inflammatory response suggesting new avenues for treatment strategies that focus on immunological targets. Despite the lack of metabolic stress induced liver phenotype, a large similarity between the NIF mouse and the immunological phase of human MASH was detected. The translational value was further confirmed through comparative analyses with MASH patients and additional animal models.
