Project description:Gene Expression dynamics is important information. To know IKK- or ERK-dependent B cell receptor- or CD40-induced gene expression dynamics, we performed the time course and dose response analysis in wild type or MEK inhibitor treated or IKKbeta inactive DT40 B cells. Two replicated samples were analysed. Unstimulated cells (T0) were control. WT cells or MEK Inh. (MEK inhibitor;U0126 5mM, 30min pretreated) or IKKbetaSA (S176/181A knock-in) Cells were stimulated with 0.1, 1, 10 Âµg/ml of anti-IgM (M4) or 1, 3, 6 Âµg/ml of CD40ligand (CD40L) for 0, 15, 30, 45, 60 or 90min.

Project description:IKKbeta is a subunit of the IkB kinase (IKK) complex required for NF-kB activation in response to pro-inflammatory signals. NF-kB regulates the expression of many genes involved in inflammation, immunity and apoptosis, and also controls cell proliferation and differentiation in different tissues; however, its function in skin physiopathology remains controversial. We here report the alterations caused by increased IKKbeta activity in basal cells of the skin of transgenic mice. Transgenic mice which express the IKKbeta gene under the control of the keratin K5 regulatory elements were generated. We analyzed back skins from wildtype and transgenic animals. Each individual array has RNA from three different mice of the same genotype.

Effect of auxin (indole-3-acetic acid) on chicken DT40 cells

Project description:DT40 cells and a stable DT 40 cell line expressing OsTIR1 (clone 9) were treated with 500µM IAA for 6 hours. This experiment was conducued to see if auxin would induce a specific sets of genes in animal, like it does for plant cells. We didn't observe a significant difference between the mock control and IAA treatments in both DT40 cells and the stable cells.

Project description:Global gene expression profiling of the avian B-lymphoma DT40 cell line was used as a model to differentiate among Btk KO and Btk KO cells reconstituted with human Btk. Differences in the gene expression pattern showed statistically significant changes between parental DT40 and all the Btk KO cell populations irrespective of whether they are reconstituted or not. These results imply that in the process of generating a knockout cell line, subclones are selected, which have multiple changes in their gene expression pattern (p<0.01). Keywords: cell line
