Project description:We investigated whether the features of donor-derived stromal vascular fraction of perirenal adipose tissue (PRAT-SVF) could be indicative of the deleterious impact of the ECD microenvironment on renal transplants. A comparative analysis of cellular components, transcriptomic and vasculogenic profiles was performed in PRAT-SVF obtained from deceased donors (non ECD) and expanded criteria donors (ECD) The global RNA sequencing approach indicated a differential molecular signature in the PRAT-SVF of ECD donors characterized by the overexpression of inflammatory transcripts (CXCL1, CCL4, IL1-β,INF-ϒ).

Suckling lamb perirenal fat transcriptome

Project description:The aim of this study was to perform a comparative analysis of the perirenal fat transcriptomes of suckling lambs from two breeds with different growth and carcass characteristics. The perirenal fat tissue from 14 suckling lambs (Assaf= 8, Churra= 6) was used for the RNA-Seq analysis. The functional enrichment analysis of the 670 highly expressed genes (>150 fragments per kilobase of exon per million fragments mapped) in the perirenal fat transcriptome of both breeds revealed that the majority of these genes were involved in energy processes. The differential expression analysis performed identified 373 differentially expressed genes (DEGs) between the two compared breeds. In Assaf lambs, DEGs were enriched in Gene Ontology (GO) biological processes related to fatty-acid oxidation, while in Churra lambs, the majority of the significantly enriched GO terms were related to cholesterol synthesis, which suggests that upregulated DEGs in Assaf lambs are implicated in fat burning, while the Churra upregulated DEGs are linked to fat accumulation.

Project description:Samples of perirenal fat tissue from 8 Assaf breed suckling lambs. These animals were selected from a larger group of 17 Assaf suckling lambs for which carcass traits were measured. The 8 selected lambs were those showing the highest and the lowest values, from the larger group, for the percentage of perirenal and cavitary fat relative to the half carcass weight. Hence, considering the values for this trait, we defined the High-PF group (n = 4; average: 3.23 ± 0,.47) and the Low-PF group (n = 4; 1.65 ± 0,.16), respectively.

Pathogenic mechanisms of Pseudomonas aeruginosa wound infection in response to murine fascia

Project description:Wound infections are traditionally thought to occur when microbial burden exceeds the innate clearance capacity of host immune system. Here we introduce the idea that the wound environment itself plays a significant contributory role to wound infection. We developed a clinically relevant murine model of soft tissue infection to explore the role of activation of microbial virulence in response to tissue factors as a mechanism by which pathogenic bacteria cause wound infections. Mice underwent abdominal skin incision and light muscle injury with a crushing forceps versus skin incision alone followed by topical inoculation of Pseudomonas aeruginosa. Pseudomonas aeruginosa whole genome transcriptional profiling demonstrated that fascia induced the activation of multiple genes responsible for the synthesis of the iron scavenging protein pyochelin. Ex-vivo murine fascia homogenates were prepared and Pseudomonas aeruginosa MPAO1 was incubated with an inoculum of the fascia homogenate solution. Pseudomonas aeruginosa MPAO1 incubated under the same condtions without the homogenate was used as the control group. Three biological replicates in each group was used.

Transcription profiling of rat Perirenal and epididymal preadipocytes isolated from old and young individuals

Project description:Inherent depot- and age-dependent preadipocyte characteristics may contribute to age-related fat redistribution. Both aging and depot origin affect preadipocyte replication and adipogenesis. To define responsible mechanisms, we analyzed genome-wide expression profiles in epididymal (E) and perirenal (P) preadipocytes cultured from young (3 month) and old (30m) rats. Differences between depots were distinct from and more dramatic than those that occur with aging. Experiment Overall Design: Preadipocytes were isolated from perirenal and epididymal fat depots of 3 young (3 month) and 3 old (30 month) Brown Norway rats for a total of 12 samples from 6 different animals.
