Project description:We have employed whole microRNA microarray to identify changes in microRNA expression in human bone marrow MSCs (hMSC-TERT) during adipocytic differentiation in culture on day 7 and day 13 Human MSC line (hMSC-TERT) were subjectd to adipocytic differentiation for 7 days or 13 days, subsequently, RNA was extracted. The samples were labeled using the miRCURY LNAM-bM-^DM-" microRNA Hi-Power Labeling Kit, Hy3M-bM-^DM-"/Hy5M-bM-^DM-" and hybridized on the miRCURY LNAM-bM-^DM-" microRNA Array (6'th GEN). Array product number 208400; Array version 6'th GEN, Array batch number 34014, miRBase version 17. The 'Legends_rawdata.pdf' contains the column headers (of raw data files) and their detailed description.

Project description:We aimed in this study to identify the differentially regulated genes by Dlk1 in hMSC cells using microarray technology in order to gain a better understanding of Dlk1-mediated signaling pathways during hMSC differentiation. Both control (hMSC-TERT)(not expressing Dlk1) and Dlk1 overexpressing cells (hMSC-Dlk1) were cultured in triplicate at 3×104 cells/cm2 in Petri-dish in standard growth medium. At 90-100% confluence, highly purified total cellular RNA was isolated from each of three independent cultures per cell line using RNeasy Kit (QIAGEN Nordic, West Sussex, UK) according to the manufacturer?s instructions

Molecular characterization of immortalization of human MSCs by a combination of p53 knockdown and TERT overexpression

Project description:Gene expression profile of p53 knockdown MSCs or p53 knockdown+TERT MSCs was compared with that of control MSCs. Our data show p53 knockdown prolongs the lifespan of MSCs, and a combination of p53 knockdown and TERT overexpression is sufficient to immortalize MSCs. The results provide important information about the molecular basis underlying p53 knockdown in MSCs and immortalization-related genes of MSCs. Total RNA obtained from p53 knockdown MSCs or p53 knockdown+TERT MSCs from three patients were compared with control MSCs.
