Gene profiling of the zone of altered morphology of choriodecidua overlying the cervix before labor in preterm samples

ABSTRACT: To characterize the molecular signature of the choriodecidua remodeling in third trimestry of pregnancy before labour, we have employed whole genome microarray expression profiling. Fetal membranes were collected from women undergoing a emergency cesarean delivery before term (before 32 weeks of gestation and between 33-35 weeks of gestation), for preeclampsia and/or in utero growth retardation. During surgery, a Kocher's forceps was placed on membranes overlying the cervix and the area surrounding the forceps was dissected in the operating room under sterile conditions, the site, pinched by the forceps, being discarded. Fetal membranes in a remote zone were also sampled. Isolated RNAs were hybridized to the Aligent SurePrint G3 Human GE 8x60K Microarray.

Project description:To characterize the molecular signature of the choriodecidua remodeling in late pregnancy befour labour, we have employed whole genome microarray expression profiling to identify genes that sign for fetal membranes remodeling. Fetal membranes were collected from healthy women, after an uncomplicated pregnancy, undergoing a planned cesarean delivery at term, for feto-pelvic disproportion or scarred uterus. During surgery, a Kocher's forceps was placed on membranes overlying the cervix and the swelling area surrounding the forceps was dissected in the operating room under sterile conditions, the site, pinched by the forceps, being discarded. Fetal membranes in a remote zone were also sampled. Isolated RNAs were hybridized to the Aligent SurePrint G3 Human GE 8x60K Microarray. Two independent bio-analyses singularize a signature in the zone of altered morphology for Graft vs Host Disease and leukocyte activation, arguing for a focal loss of maternal-fetal tolerance. This was further confirmed by flow cytometry, electron microscopy, and immuhistochemistry analyses.

Large-scale downregulation of cytokeratins in amniotic epithelium after Group B Streptococcus choriodecidual infection in Macaca nemestrina: a pathway to PPROM

Project description:Early events leading to intrauterine infection remain poorly defined, but may hold the key to a therapeutic intervention for preterm delivery. To determine early molecular pathways associated with membrane weakening that may progress to preterm premature rupture of membranes (PPROM), we examined the effects of Group B Streptococcus (GBS) on fetal membranes after a choriodecidual infection. Ten chronically catheterized pregnant monkeys (Macaca nemestrina) at 118-125 days gestation (term=172 days) received choriodecidual inoculation of either: 1) Group B Streptococcus (n=5) or 2) saline (n=5). Cesarean section and fetal necropsy was performed in the first week after GBS or saline inoculation regardless of labor. Macaca nemestrina chorioamnion samples were obtained at the site of inocculation immediately after cesarean delivery 4 days after GBS inoculation and 7 days after saline inoculation. RNA was extracted and profiled by microarray. Results were analyzed using single gene, Gene Set, and Ingenuity Pathway Analysis. Validation was by RT-PCR and immunohistochemistry.

Data in support of Prenatal intra-amniotic lipopolysaccharide induces neonatal renal immune activation after preterm birth

Project description:Preterm birth is often predisposed by chorioamnionitis (CA) and CA affects the fetal gut and lungs via intra-amniotic (IA) inflammation, thus accentuating the proinflammatory effects of preterm birth. It is not known if IA inflammation also affects other perfusion-sensitive organs (e.g., kidneys) before and after preterm birth. Using preterm pigs as model for preterm infants, we hypothesized that CA induces fetal and neonatal renal dysfunctions that can intially be detected via plasma proteome, partly explaining the frequent renal dysfunction in preterm infants. Fetal pigs (88% gestation) were given an IA dose of lipopolysaccharide (LPS, 1 mg/kg, n=28), delivered preterm by cesarean section three days later, and compared with controls (CON, n=26) at birth and postnatal day five. Plasma proteome and protein markers of inflammatory pathways were evaluated.
