ABSTRACT: Transcriptional profiling of Acinetobacter baumannii ATCC17978 cells comparing treated ethanol cells with oleanolic acid treated. Based on the gene expression, we performed experiments to confirm the therapeutic effect and mechanism of OA in A. baumannii.

Differentially regulted genes in neural stem cells treated by oleanolic acid

Project description:Oleanolic acid significantly inhibited neurosphere formation in a dose-dependent manner, and achieved a maximum effect at 10 nM. OA also reduced the 5-ethynyl-2M-bM-^@M-^Y-deoxyuridine (EdU) incorporation into NSCs, indicating an inhibition on proliferation of NSCs. Next, in western blotting analysis, the protein expression of neuron-specific marker tubulin-M-NM-2M-bM-^EM-" (TuJ1) and Mash1 was increased, while the astrocyte-specific marker glial fibrillary acidic protein (GFAP) decreased. In immunofluorescence analysis, OA significantly elevated the percentage of TuJ1-positive cells and reduced GFAP-positive cells. Using DNA microarrays, 183 genes were found to be differentially regulated by OA. Neural stem cells derived form mouse embrynic brains were treated with Oleanolic acid or not. Each group had 4 biological relicates each from a mouse.

Comparison of the gene expression profiles of different strains of A. baumannii by microarrays

Project description:In the present work we compare the gene expression profile of A. baumannii and a mutant knock-out strain of A. baumannii lacking a small RNA gene 13573 and the corresponding small RNA 13573 over-producing strain. The main objective is to recognize the main pathways in which the small RNA 13573 is involved. Moreover, the same wild type strain was used to infect mice and was further analyzed after the infection with the aim of finding genes differentially expressed in vivo. Three biological replicates have been performed for each comparison. The RNA collection from Acinetobacter baumannii strain over-expresing the small RNA (sample 13573) was compared with this isolated from A. baumannii harboring the empty vector (PETRA sample) while gene expression in the knock-out strain (KO sample) was compared with the wild type strain Acinetobacter baumannii ATCC 17978 (ATCC sample). The RNA from A.baumannii recovered from the infected animals (INF sample) was compared with the wild type (ATCC).
