Gene Expression Time Courses of CH999 with High or Low Copy Number Plasmids

ABSTRACT: Time courses of Streptomyces CH999/pKOS011-26 and CH999/pKOS011-26* the low and high copy number strains, respectively, to examine gene expression differences between these two strains. Of particular interest were the transcript levels of the activator gene actII-ORF4 and the polyketide synthase gene cluster eryA. Mycelia were grown in 50 mL R5 liquid medium with thiostrepton drug selection at 50 micrograms per mL in a 30 degrees Celsius shaker incubator. mRNA were harvested at the time points indicated. These data show that actII-ORF4 and eryA transcripts were more highly expressed in the high copy number strain than in the low copy number strain. Transcript-level responses of Plasmodium falciparum to thiostrepton

Project description:Abstract: The antimalarial activity of the antibiotic thiostrepton has long been attributed to inhibition of apicoplast protein synthesis through binding of apicoplast ribosomal RNA. However, the kinetics of parasite death upon thiostrepton treatment differ from those seen for other inhibitors of apicoplast housekeeping functions. We have analysed global changes in gene expression of the malaria parasite, Plasmodium falciparum, in an attempt to shed light on the responses of the parasite to this drug. Our results indicate a delay in gene expression profiles of thiostrepton-treated parasites. A small number of genes appear to be regulated outside of this trend; our data suggest a response from genes encoding components of the mitochondrial translational machinery, while little response is seen from genes encoding apicoplast-targeted proteins. Our findings are consistent with an effect of thiostrepton on mitochondrial protein synthesis, and thus warrant a re-evaluation of the target of thiostrepton in Plasmodium. They also provide some suggestion of mitochondrion – nucleus signalling in the parasite.

Gene expression data of nasopharyngeal carcinoma (NPC) cell and Thiostrepton treatment

Project description:Thiostrepton, a natural antibiotic, has recently been shown to be a potential anticancer drug for certain cancers, but its study in nasopharyngeal carcinoma (NPC) is still limited. The aims of this study were to investigate the anticancer effect of thiostrepton on NPC cells and to explore its underlying mechanism. Our results showed that thiostrepton reduced NPC cell viability in a dose-dependent manner. Thiostrepton inhibited the migration and invasion of NPC cells in wound healing and cell invasion assays. Thiostrepton effectively suppressed NPC cell proliferation, migration, and invasion, likely by several mechanisms. Thiostrepton may be a potential therapeutic agent for treating NPC in the future.
