Project description:We have used small RNA sequencing to measure the expression of all miRNAs present in the kidneys during the development of folic acid-induced kidney injury and fibrosis. From this miRNA expression profile, we identified miRNAs associated with different stages and aspects of kidney damage. BALB/c mice were injected with 250mg/kg of folic acid and kidneys were collected at 0, 1, 2, 3, 7, 14, and 28 days. Total RNA was prepared for sequencing using the Illumina TruSeq Small RNA Sample Preparation Kit, and analyzed on an Illumina HiSeq 2000.

Project description:Treatments for kidney fibrosis represent an urgent yet unmet clinical need. Effective therapies are limited due to not well understood molecular pathogenesis. We aimed at generating a comprehensive and integrated multi-omics data set (RNA/ microRNA transcriptomics and proteomics) of fibrotic kidneys which will be searchable through a user-friendly web application. Therefore, two commonly used mouse models were utilized: a reversible chemical-induced injury model (folic acid (FA) induced nephropathy) and an irreversible surgical-induced fibrosis model (unilateral ureteral obstruction (UUO)). RNA and small RNA sequencing as well as MS/MS with 10-plex tandem mass tags proteomics were performed with kidney samples from different time points over the course of fibrosis development. In summary, we present temporal and integrated multi-omics data from fibrotic mouse kidneys which are accessible through an interrogation tool to provide a searchable transcriptome and proteome for kidney fibrosis researcher.

Two commonly used mouse models were utilized: a reversible chemical-induced injury model (folic acid (FA) induced nephropathy) and an irreversible surgically-induced fibrosis model (unilateral ureteral obstruction (UUO)). mRNA and small RNA sequencing as well as 10-plex tandem mass tag (TMT) proteomics were performed with kidney samples from different time points over the course of fibrosis development. The bioinformatics workflow used to process, technically validate, and integrate the single data sets will be described. In summary, we present temporal and integrated multi-omics data from fibrotic mouse kidneys that are accessible through an interrogation tool to provide a searchable transcriptome and proteome for kidney fibrosis researchers.

Project description:To investigate altered expression patterns of key miRNAs in end-stage ADPKD kidneys, a miRNA microarray analysis was performed using non-ADPKD and ADPKD kidney tissue samples. As a result, total 19 miRNAs were significantly changed in the ADPKD samples compared with non-ADPKD samples. Among these miRNAs, the expression level of the miR-192 family including miR-192 and miR-194 were significantly downregulated in ADPKD.

Project description:Neural tube defects (NTDs) are a common and devastating birth defect. The current NTD rate in the U.S. reflects an approximately 30% decrease following folic acid (FA) supplementation in the 1990’s as a public health measure. Yet, questions remain including which gene mutations are sensitive to folic acid levels and how folic acid acts to alter NTD risk. We studied the relationship between FA dosage and NTD rates in NTD mouse models and propose that genes involved in cilia formation and function represent an FA sensitive ontology and a possible avenue for further reducing NTD rates. We demonstrate that in cilia mutant mice, a more moderate level of FA is beneficial in reducing NTD rates and promoting primary and motile cilia formation and function. We present a mechanism through which fortified FA levels reduce basal levels of reactive oxygen species (ROS) which in turn reduces ROS-sensitive GTPase activity required for ciliogenesis.

Project description:The microRNAs (miRNAs) that can influence diabetic kidney disease (DKD) have not been fully characterized. The aim of this study was to identify the miRNAs that affect DKD and could be used as specific biomarkers or therapeutic agents. First, kidneys from two DKD mouse models were screened for differences in miRNA expression from control mice. We found that miRNA-125b-5p and miRNA-181-5p were specifically differentially expressed in the kidneys of DKD mice. Next, we administered miRNA-181b-5p-mimic to DKD mice, which reduced the albuminuria and abnormal mesangial expansion. Pathway analysis revealed that overexpression of miRNA-181-5p significantly altered the expression of 51 genes in the kidneys of DKD mice. Furthermore, the serum level of miRNA-125b-5p was significantly higher and that of miRNA-181-5p was lower in patients with DKD than in patients with other kidney diseases. These results suggest that miRNA-125b-5p and miRNA-181b-5p may represent novel diagnostic biomarkers and that miRNA-181b-5p may represent a therapeutic target for DKD.
