Oxi1 transcriptome-Transcriptomic analysis on the effect of oxi1 mutation under high light.

ABSTRACT: cea13-01_oxi1 - oxi1 transcriptome - Transcriptomic analysis on the effect of oxi1 mutation under high light stress? - 5 weeks old mutannt (M) and wild type (WT) plants were exposed to high light and low temperature (1300-1350 µmol photons m-2s-1, 7°C/14°C day/night and 380ppm CO2) for 25 hours. ~100 mg fresh weight leaves were harvested, and total RNA prepared from them. For control experiments, leaves were harvested directly from the phytotron (No light stress). Three microarray comparisons were made: Mcontrol/WTcontrol, Mstress/WTstress and Mstress/Mcontrol . For each comparison, 2 biological replicates are performed.

RNA-seq of etiolated 9 day old maize seedling second leaves after exposure to either blue, red or no light conditions for three hours

Project description:Characterisation of the differences in transcript abundance between maize leaves that have been exposed to blue, red, or no light: B73 maize seeds were planted and grown in the dark for 9 days. Etiolated second leaves were clamped 5 cm from the leaf tip by a Licor 6800 device equipped with a multi-phase flash fluorometer head, which administered 100 µmol m-2s-1 of either 100% red or 100% blue light. The Licor was configured with flow rate 500 µmol s⁻¹, 400 µmol mol⁻¹ CO2, leaf temperature 28°C and 60% humidity. Fluorescence and gas exchange were measured every 15 minutes for 3 hours. Leaf samples were collected between 12.30am-2pm each day.

RL-SAGE analysis of Zea mays leaf transcriptome

Project description:Using the RL-SAGE method (Gowda et al. 2004), a maize leaf longSAGE library (cv. inbred line B73) was constructed. Leaf tissues were harvested from 4-week old B73 plants for RNA isolation. The conditions in the growth chamber were 12 h light (500 µmol photons m-2 sec-1), 20oC at night, 26oC in the day and 85% relative humidity. A total of 44,870 unique tags (17 bases +CATG) were identified from 232,948 individual tags in the maize leaf library.

Carbon fixation (endogenous sugar) and light-dependent gene expression

Project description:Internal sugar and light specific dependent regulation of leaf gene expression was addressed by changing [CO2] to lower than compensation point [CO2] in combination with light or prolonged darkness. Plants were grown on soil in a 12/12 h light/dark rhythm at 20°C day and night and under normal [CO2]. 5 weeks after germination, the above-ground rosettes of the non-flowering plants were harvested, 12 plants per sample. Plants were harvested 4hrs after the end of night (i) under low (< 50 ppm) [CO2] and 150 µE fluorescent light , (ii) under normal [CO2] and light, and, (iii) under low [CO2] and prolonged darkness. Synechocystis sp. proteome on differnt light conditions

Project description:Cyanobacteria are an integral part of the Earth’s biogeochemical cycles and a promising resource for the synthesis of renewable bioproducts from atmospheric CO2. Sustainable industrial applications, however, are still scarce and the true limits of phototrophic production remain unknown. One of the limitations of further progress is our insufficient understanding of the quantitative changes in photoautotrophic metabolism that occur during growth in dynamic environments. Uncovering principles of allocation of the key metabolites or proteins as a response to changing environment can provide significant advances in our understanding of phototrophic production limits. However, resolving such complex question requires multidisciplinary approach combining highly-controlled cyanobacteria cultivation coupled to quantitative analyses of specific cellular components. The following datase represents proteomic analysis of the model cyanobacterium Synechocystis sp. PCC 6803 that was cultivated in the well controlled environment of laboratory-scale photobioreactors in the quasi-continuous regime that allowed for maintaining the cells in the exponential growth phase. The sampels were taken under light-limited (25 - 220 µmol(photons) m-2 s-1), optimal (440 µmol(photons) m-2 s-1) and photoinhibited growth (1100 µmol(photons) m-2 s-1). Each culture was sampled in five biological replicates.

Leaves were harvested at three different time points: t = 0 hr (end of night), t = 1 hr (one hour after light turn on) and t = 8 hr (eight hours after light turn on), respectively.

RNA-seq of an adult Porites rus colony derived from Indonesia

Project description:An adult P. rus colony was imported from Indonesia following the CITES protocols (Permit number 14846/IV/SATS-LN/2007) and kept in the Animal Facilities of the Justus Liebig University, Giessen. The colony was maintained in a circulating artificial seawater system at approximately 26 °C with 20-40 µmol photons m-2s-1 (T5 light) of photosynthetically active radiation on a 10:14 h light-dark cycle. A fragment of approx. 9 cm2 was separated from the colony and used as a source of tissue for hologenomic DNA/RNA isolation. Tissue was removed by scraping the fragment´s surface with a sterilized razor blade.
