Identifying the targets of the pair-rule transcription factors Even-skipped and Runt in the late Drosophila blastoderm

ABSTRACT: It has long been appreciated that striped pair-rule transcription factor expression is necessary for convergent extension in the early Drosophila embryo, although the mechanisms that link these transcriptional regulators to planar polarity in this tissue have long been elusive. The goal of this study was to determine the transcriptional tragets of the pair-rule transcription factors Eve and Runt in Drosophila blastoderm embryos. We compared the transcriptional profiles of late blastoderm embryos injected with either water or dsRNAs against both eve and runt to identify differentially expressed genes that may directly contribute to the establishment of planar polarity during Drosophila convergent extension.

A novel Golgi-associated Vangl2 Translational variant is required for PCP regulation in vertebrates

Project description:First described in Drosophila melanogaster, planar cell polarity (PCP) is a developmental process essential for embryogenesis and development of polarized structures in Metazoans. This signaling pathway involves a set of evolutionarily conserved genes encoding transmembrane (Vangl, Frizzled, Celsr) and cytoplasmic (Prickle, Dishevelled) molecules. Vangl2 is of major importance in embryonic development as illustrated by its pivotal role during neural tube closure in human, mouse, Xenopus and zebrafish embryos. The regulated and poorly understood traffic of Vangl2 to the plasma membrane is a key event for its function in development. Here we identify a novel N-terminally extended isoform of Vangl2, termed Vangl2-Long that arises from the use of non-AUG start codon upstream of the coding region of canonical Vangl2. Vangl2-Long contains an evolutionarily conserved N-terminal 48 amino acid sequence bearing a signal for subcellular localization in the Golgi apparatus. Moreover, we provide data in Xenopus showing that Vangl2-Long is important for correct convergent extension movements and neural tube closure. These data describe a further level of complexity in Vangl2 expression, trafficking and function.

Early developmental transcriptome of Musca domestica.

Project description:Using mRNA next generation sequencing, we analyze the transcriptome of Musca domestica embryos through five stages of early development: from syncytial blastoderm to dorsal closure stage. We select 100 embryos for five developmental stages: syncytial blastoderm, cellular blastoderm, gastrula, germ band extension and dorsal closure. A total of 10 samples of mRNA were obtained, represent two biological replicates for each stage.

ChIP-seq of Atrophin in Drosophila S2 cells

Although Atro mutants have diverse phenotypes, little is known about Atro’s binding partners and downstream targets. Project description:The orphan nuclear receptor Ftz-F1 is expressed in all somatic nuclei in Drosophila embryos but mutations result in a pair-rule phenotype. Previously characterized Ftz-F1 target genes were co-regulated by Ftz, which is expressed in stripes, consistent with the pair-rule phenotype observed for ftz-f1 and ftz mutants. However, attempts to identify new target genes on the basis of Ftz-F1 and Ftz binding sites alone has met with only limited success. To discern the rules for Ftz-F1 target site selection in vivo and to identify additional target genes, a microarray analysis was performed comparing wildtype and ftz-f1 mutant embryos.
