Comparative Genomic Hybridizaions of 3D7∆PfSir2a and NF54 compared to reference strain 3D7

ABSTRACT: Antigenic variation in Plasmodium falciparum is achieved by transcriptional switching amongst polymorphic var genes, enforced by epigenetic modification of chromatin. Histone-modifying ‘sirtuin’ enzymes PfSir2a and PfSir2b have been implicated in this process. We examined the effect of genetic disruption of sirtuins on var gene expression. Comparative Genomic Hybridization profile indicate that loss of PfSir2a in 3D7 resulted in strikingly rearranged chromosomes.

3D7 PfHda2 Knockdown vs. Wildtype

Project description:The asexual forms of the malaria parasite Plasmodium falciparum are uniquely adapted for chronic persistence in human red blood cells, continuously evading the immune system using an epigenetically regulated process. However, parasite survival on a population level also requires transmission of sexual parasite forms to subsequent human hosts. Here, we reveal that the essential nuclear gene, P. falciparum histone deacetylase 2 (PfHda2), silences specific subsets of genes involved in antigenic variation or conversion to sexual stages. Two parallel timecourses resulting in a total of 22 samples (11 wildtype, 11 PfHda2 knockdown) were hybridized against a Cy3-labeled reference pool of 3D7 mixed stage parasites on a two-color array.

Specific functions for the fission yeast Sirtuins Hst2 and Hst4 in gene regulation and retrotransposon silencing

Project description:Expression profiling, ChiP-CHIP and phenotypic analysis were used to investigate the functional relationships of class III NAD+-dependent HDACs (Sirtuins) in fission yeast. We detected significant histone acetylation increases in Sirtuin mutants at their specific genomic binding targets and were thus able to identify an in vivo substrate preference for each Sirtuin. At heterochromatic loci, we demonstrate that although Hst2 is mainly cytoplasmic, a nuclear pool of Hst2 co-localises with the other Sirtuins at silent regions (cen, mat, tel, rDNA) and that like the other Sirtuins, Hst2 is required for rDNA and centromeric silencing. Interestingly we found specific functions for the fission yeast Sirtuins Hst2 and Hst4 in gene regulation. Hst2 directly represses genes involved in transport and membrane function whereas Hst4 represses amino acid biosynthesis genes and Tf2 retrotransposons. A specific role for Hst4 in Tf2 5’ mRNA processing was revealed. Thus, Sirtuins share functions at many genomic targets, but Hst2 and Hst4 have also evolved unique functions in gene regulation. Sustained activation of AMPK enhances differentiation of human iPSC-derived cardiomyocytes via sirtuin activation

Project description:Lee and colleagues demonstrate that sustained activation of AMPK enhances differentiation of iPSC-derived cardiomyocytes. Sustained AMPK activation decreased histone acetylation at known target sites for nuclear-localized sirtuins, suggesting that AMPK activation enhances sirtuin activity. AMPK-induced sirtuin-mediated deacetylation of histone proteins may regulate chromatin accessibility and enhance cardiomyocyte differentiation.

Transcriptional alterations of P. falciparum 3D7-A heat shock-adapted vs control parasites.

Project description:Periodic fever is the most characteristic clinical feature of human malaria. However, how parasites survive malarial febrile episodes, which often involve temperatures of >40ºC, is not known. To understand the molecular basis of heat shock (HS) resistance in Plasmodium falciparum, we took advantage of previously developed P. falciparum lines adapted to periodic HS (3D7-A-HS) and their non-selected controls maintained in parallel (3D7-A). Their ability to adapt in only a few cycles suggested that the parental parasite population contained a selectable subset of parasites resistant to HS. Transcriptomic analysis of both 3D7-A-HS and 3D7-A was used to study the differences at a transcriptional level that could be related with the HS-resistant phenotype.
