Time course microarray analysis during xylem cell differentiation in leaf-disk culture system

ABSTRACT: We established a novel in vitro culture system, in which vascular development can be induced in Arabidopsis thaliana leaf-disks using the specific GSK3s inhibitor. To elucidate gene expression profiles during vascular development, we performed GeneChip analysis using our newly established culture system.

CC-M means a sample that moderately induces CC differentiation. V means a VISUAL sample, that does not induce CC differentiation at all.

Analysis of wound-induced transcripts in poplar leaf disks versus whole leaves

Project description:Expression of defense response-related transcripts could be affected upon leaf wounding such as after excision of leaf disks. In this experiment, transcripts were isolated from whole leaves and 5-cm2 excised leaf disks samples from interaction assays between leaves of the 'Beaupré' poplar cultivar (Poplus trichocarpa x Poplus deltoides) and virulent (98AG31, compatible interaction) and avirulent (93ID6, incompatible interaction) urediniospores of Melampsora larici-populina, the fungus responsible for the foliar rust disease in poplar. For each type of interaction, three samples were collected in a time-course series (18, 21 and 24 hpi for strain 93ID6, and 18, 24 and 48 hpi for strain 98AG31). Both leaf disks and whole leaves were inoculated in strictly similar conditions, so differential transcript expression is likely attributable to the wounding effect in leaf disks. The aim of this study is to identify genes that are induced by wounding in leaf disks compared with whole leaves. For this purpose, cDNA templates were used for hybridization on poplar oligoarrays.

Global gene expression changes including drug metabolism and disposition induced by three-dimensional culture of HepG2

Project description:We found constitutive upregulation and higher degree induction of drug metabolism and disposition-related genes in a three-dimensional HepG2 culture. The upregulated genes are those believed to be regulated by different regulatory factors. The global gene expression analysis by Affymetrix GeneChip indicated that altered expressions of microtubule-related genes may change expressed levels of drug metabolism and disposition genes. Stabilization of the microtubule molecules with docetaxel, a tubulin stabilizing agent, in the two-dimensional culture showed gene expression patterns similar to those in the three-dimensional culture, indicating that culture environment affects drug metabolism functions in HepG2 cells.
