Expression profile from Saccharomyces cerevisiae strains deleted for PMR1 treated with 5mM CaCl2

ABSTRACT: Pmr1 is a cis-Golgi Mn/Ca transporter with a key role in protein glycosylation and manganese detoxification. We used microarray analysis to analyze the impact of CaCl2 supplementation on gene expression in pmr1∆ mutants and found that extracellular CaCl2 suppresses transcriptional misregulation in pmr∆ mutants.

Project description:The yeast PMR1 (ATP2C1) gene codes for the eukaryotic prototype of a high affinity P-type ATPase required for Ca2+/Mn2+ transport into the Golgi. Cells lacking PMR1 exhibit multiple genetic interactions with genes involved in DNA recombination and replication, a fact that is not yet understood. We find that deletion of PMR1 causes a delay in DNA replication initiation, progression and G2/M transition and induces the transcriptional up-regulation of genes involved in cell cycle regulation, including CLB5 and SWE1. Interestingly, pmr1∆ clb5∆ double mutants exhibit a dramatic delay in DNA replication and increased DNA breakage, while endoreplication and the formation of multi-nucleated, giant yeast is observed in pmr1∆ swe1∆ cells. Because these phenotypes can be attributed to impeded Mn2+-pump function, we provide a model in which Mn2+ interferes with Mg2+ in the nucleus, and vice versa, Mg2+ interferes with Mn2+ in the Golgi. Consequently, cell cycle progression is challenged by aberrant catalytic activities of enzymes involved in replication and protein glycosylation.

Transcript levels of genes in the wild type CAI4 as well as the gdt1/gdt1,the pmr1/pmr1 and the gdt1/gdt1 pmr1/pmr1 mutants

Project description:We report the expression levels of transcripts of 5,901 genes in log-phase growing cells of the wild type CAI4 as well as the gdt1/gdt1, the pmr1/pmr1, and the gdt1/gdt1 pmr1/pmr1 mutants.We find that as compared to the wild type, there are 249, 446 and 440 differentially expressed genes (DEGs) in the gdt1/gdt1, the pmr1/pmr1 and the gdt1/gdt1 pmr1/pmr1 mutants, respectively. Deletion of CaGDT1 does not affect the transcript level of CaPMR1, and vice versa. There are 105 shared DEGs between the gdt1/gdt1 and the pmr1/pmr1 mutants, which accounts for 42% (105/249) of total DEGs due to deletion of CaGDT1 or 24% (105/446) of total DEGs due to deletion of CaPMR1.

The hierarchy of transition metal homeostasis: Iron controls manganese accumulation in a unicellular cyanobacterium.

Project description:Iron and manganese are part of a small group of transition metals required for photosynthetic electron transport. Here, we present evidence for a functional link between iron and manganese homeostasis. In the unicellular cyanobacterium, Synechocystis sp. PCC 6803 Fe and Mn deprivation resulted in distinct modifications of the function of the photosynthetic apparatus. For example, iron limitation modifies the rate of QA re-oxidation in photosystem II, a complex that contains more Mn than Fe. The intracellular elemental quotas of Fe and Mn are also linked. Fe limitation reduces the intracellular Mn quota. Mn limitation did not exert a reciprocal effect on Fe quotas. Microarray analysis comparing Mn and Fe limitation revealed a stark difference in the extent of the transcriptional response to the two limiting conditions, reflective of the physiological data. The effects of Fe limitation on the transcriptional network are widespread while the effects on Mn limitation are highly specific. Our analysis also revealed an overlap in the transcriptional response of specific Fe and Mn transporters. This overlap provides a framework for explaining Fe limitation induced changes in Mn quotas. Fe transporters can serve as a low affinity Mn transport system. Under iron limitation the specificity of the Fe transport system changes, making it a less efficient Mn transport system. We monitored the gene expression of Synechocystis PCC6083 at standard conditions and after 2 days of iron limitation (0Fe), manganese limitation (0Mn) and combined iron and manganese limitation (0Fe0Mn). Each timepoint and condition was sampled in triplicates. Due to strong deviations in one of the three repeats for the 0Mn and 0Fe0Mn conditions, the corresponding replicates were excluded from further analysis.

Project description:The hypothesis whether CaCl2 seed treatment prior to sowing (osmopriming) can enhance drought stress tolerance, and/or alleviate the effect on plant growth was tested. Here we elucidate whether seed treatment prior to sowing (osmopriming) with CaCl2 can improve the drought stress tolerance, as it was previously reported for salt stress on wheat. Thus the effect of 50 mM CaCl2, previously chosen based on pilot experiment, versus water-treated control exposed to drought stress was investigated.

Transcript levels of genes in both the wild type SN148 and the crz1/crz1 mutant with or without 0.2M CaCl2 treatment

Project description:We report the expression levels of transcripts for two alleles of 6,211 genes in log-phase growing cells of the wild type SN148 and the crz1/crz1 with or without 0.2M CaCl2 treatment.We find that as compared to the wild type cells without 0.2M CaCl2 treatment, there are 828 genes upregulated in the wild type cells with 0.2M CaCl2 treatment.
