Project description:Hypertension is a major risk factor for chronic kidney disease (CKD) and renal inflammation is an integral part in this pathology. Hydrogen sulfide (H2S) has been shown to mitigate renal damage through reduction in blood pressure and reactive oxygen species; however, the exact mechanisms are not clear. While several studies have underlined the role of epigenetics in renal inflammation and dysfunction, the mechanisms through which epigenetic regulators play role in hypertension are not well defined. We used microarrays to detail the global programme of gene expression underlying hypertension in the kidney and how hydrogen sulfide supplementation alleviates these effects.

Project description:Many diseases, including diabetes and hypertension, can lead to kidney damage, often resulting in long-term health complications and organ failure. Both animal and human studies have documented a causal relationship between high dietary sodium intake and elevated blood pressure. We explored a proteomics analysis of plasma samples to detect early kidney damage in response to a high sodium (HS) diet through the quantification of putative kidney-derived proteins in plasma for early disease detection. Plasma samples from 7 female baboons fed a high sodium diet for 6 weeks were collected before and after the HS diet challenge. Plasma samples were analyzed using a novel nanoparticle enrichment protocol.

Project description:Inflammation and sympathetic overactivity contribute to end-organ injury associated with hypertension. We previously demonstrated that targeting formylpeptide receptors (FPRs), which regulate and resolve inflammation, prevents cardiovascular damage in preclinical hypertension. Here, we investigated whether the FPR agonist Cmpd17b can reverse renovascular remodeling in established essential hypertension. Cmpd17b selectively reduced blood pressure in BPH/2J mice and improved aortic distensibility without altering cardiac function. Cmpd17b reduced collagen deposition across organs, most prominently in the kidney. RNA sequencing revealed kidney-specific upregulation of Serpina1e and Igfbp2, normalized by Cmpd17b. These findings identify FPR agonism as a promising therapeutic strategy for lowering blood pressure and reversing renovascular fibrosis.

Project description:The left kidneys of 8 Sprague-Dawley rats were removed and the rats were left for 2 weeks to recover. After 4 additional days (during which blood pressure measurements were obtained), they were injected daily subcut. with either saline (1ml/kg), adrenocorticotropic hormone (ACTH, 0.2mg/kg) or dexamethasone (DEX, 0.02mg/kg) for 8 days before the right kidney was removed. ACTH and DEX treatment caused hypertension in the rats as illustrated by increased systolic blood pressure. The gene expression patterns of the kidneys were analysed using Affymetrix arrays. Experiment Overall Design: The left kidney of each rat acts as a control for the treated right kidney. Two rats each were treated with either saline, ACTH or DEX.

Project description:Aldosterone is arguably the single most important hormone implicated in the control of blood pressure and extracellular fluid volume in mammals. It acts primarily by increasing the rate of transepithelial sodium transport in the kidney tubules. Several monogenetic defects resulting in hypertension have now been attributed to abnormalities in sodium handling within the aldosterone-sensitive distal nephron, where the epithelial sodium channel, ENaC, constitutes the rate-limiting step of sodium transport. To date, the transcription-dependent aldosterone-signaling pathway between receptor and membrane transport effectors, remains incompletely understood. The broad aim of our study is to explore these intracellular signaling pathways that are critical to the functioning of sodium channels. Microarray analysis in an aldosterone-responsive kidney cortical collecting duct cell line (mpkCCDc14), allowed us to identify many potential aldosterone-regulated transcripts. The present study describes the identification, and possible physiological relevance of various aldosterone-regulated transcripts. The results of this study promise to extend our understanding of the molecular basis of aldosterone-regulated sodium transport in kidney epithelia, and provide approaches for regulating this process in disease states such as congestive heart failure and hypertension. Age-related differences in kidney biology, therefore, are a key concern in understanding drug safety and disease progression. We hypothesize that the underlying suite of genes expressed in the kidney at various life cycle stages will impact susceptibility to adverse drug reactions. Therefore, establishing changes in baseline expression data between these life stages is the first and necessary step in evaluating this hypothesis.

Project description:Chronic Kidney Disease (CKD), a global health burden, is strongly associated with age-related renal function decline, hypertension and diabetes, frequent consequences of obesity. Despite extensive studies the mechanisms determining susceptibility to CKD remain elusive. Clinical evidence together with prior studies from our group showed that perinatal metabolic disorders after maternal obesity adversely affect kidney structure and function throughout life. Since obesity and aging processes converge in similar pathways we tested if perinatal obesity induced by High-Fat Diet (HFD)-fed female mice sensitizes aging-associated mechanism in kidneys of newborn mice. Tissue from the kidney cortex and the kidney medulla was collected from offspring of control or HFD-fed mice.

Project description:Hypertension is a progressive condition that predisposes patients to the development of chronic kidney disease (CKD). Previous genetic and animal studies utilizing the Dahl salt-sensitive (SS) rat, a genetic model of progressive hypertension and CKD, identified that Arhgef11 was playing a role in hypertension and CKD in the Dahl SS model. Arhgef11 is a Rho guanine nucleotide exchange factor involved in the regulation of the cytoskeleton, intracellular trafficking, protein-protein interactions, and transcription factor regulation. Temporal evaluation of SS-WT and SS-Arhgef11-/- rats demonstrated that the loss of Arhgef11 attenuated hypertension and improved renal function. Transcriptome and proteomic studies lead to the hypothesis that dysregulation of these pathways in the kidney, particularly the proximal tubule, may alter sodium handling and ultimately lead to hypertension. Current rat studies confirmed previous work between SS-WT and SS-Arhgef11-/- and investigated early (week 4) transcriptional changes using single nuclei RNA sequencing, present before blood pressure differences are observed. Due to only being able to test systemic loss of Arhgef11 in the rat model, a global knockout (Arhgef11-/-(CMV)), as well as a proximal tubule-specific deletion (Arhgef11-/-(Sglt2)) were studied in the mouse. Studies utilized two models of experimental hypertension: Angiotensin II and deoxycorticosterone acetate (DOCA) + salt. Both models demonstrated a slight protective effect on blood pressure and kidney injury for DOCA experimental models, replicating the rat knockout data. However, the impact of the loss of Arhgef11 was dependent on the type of induced hypertension, with the large effect in the rat model likely explained by the highly permissive genetic background of the Dahl SS model.
