Project description:To understand plant adaptation to heat stress, gene expression profiles of Arabidopsis leaves under heat stress, during recovery and control condition were obtained using microarray. Microarray data listed responsible candidate genes for glycerolipid metabolism. Arabidopsis thaliana ecotype Columbia (Col-0) seeds were surface-sterilised and sown on an agar-solidified Murashige and Skoog medium. Plants were grown at 22ºC under a 16-h-light/8-h-dark cycle. Vegetative plants were subjected to high temperature for a day, then continued to be grown under normal condition for a day.

Transcriptomic analysis during heat stress and after the following recovery in grapevine (Vitis vinifera L.) leaves

Project description:To elucidate the effect of heat stress and the following recovery on grapevines and identify some regulated genes representing the classical heat stress response and thermotolerance mechanisms, transcript abundance of grapevine (Vitis vinifera L.) were quantified using the Affymetrix Grape Genome oligonucleotide microarray (15,700 transcripts), followed by quantitive Real-Time PCR validation for some transcript profiles. Time series of gene expression profiles from C. elegans exposed to heat-stress and during subsequent recovery

Project description:This data set provides insight into the temporal dynamics of gene expression changes during heat-stress (at 35C) and during subsequent recovery (at 20C) in Caenorhabditis elegans. Age synchronized C. elegans CB4856 hermaphrodite populations were exposed to heat stress (35C) for 0h, 0.5h, 1h, 2h, 4h, and 6h. A recovery time-series was created by using age-synchronized N2 populations exposed to heat stress (35C) for 2 hours with a subsequent recovery period of 0.5h, 1h, 1.5h, 2h, 4h, and 6h at 20C. The samples were collected at the indicated time-points during heat stress or recovery. Gene expression profiles were created using microarray technology. This data set is an extension of the data provided under accession number E-MTAB-5753 (i.e. time series describing the gene expression dynamics during heat stress of N2).

Long recovery after heat shock

Project description:Abiotic stress is a major factor for crop productivity, a problem likely to be exacerbated by climate change. Improving the tolerance to environmental stress is one of the most important goals of crop breeding programmes. While the early responses to abiotic stress in plants are well studied, plant adaptation to enduring or recurring stress conditions has received little attention. This project investigates the molecular mechanism of the maintenance of acquired thermotolerance as a model case of stress memory in Arabidopsis. Arabidopsis seedlings acquire thermotolerance through a heat treatment at sublethal temperatures. To investigate the underlying mechanisms, we are investigating changes in the transcriptome at two timepoints after a heat acclimation treatment using Arabidopsis thaliana seedlings. Microarrays were used to compare gene expression at two timepoints after a heat acclimation treatment.

A census of Hsp70-mediated proteome solubility changes upon recovery from severe heat stress

Project description:Eukaryotic cells respond to heat shock through several regulatory processes including upregulation of stress responsive chaperones and reversible shutdown cellular activities through formation of protein assemblies. However, the underlying regulatory mechanisms of the recovery of these heat-induced protein assemblies remain largely elusive. Here, we measured the proteome abundance and solubility changes during recovery from severe heat shock in the mouse Neuro2a cell line. We found that prefoldins and translation machinery are rapidly down-regulated as the first step in the heat shock response. Analysis of proteome solubility reveals a rapid mobilization of protein quality control machineries, changes in cellular energy metabolism, changes in translational activity and nucleocytoplasmic transport are fundamental to the early stress responses, while the longer term adaptation to stress involves renewal of core cellular components. Hsp70 inhibition negatively affects the ribosomal machinery and delays the solubility recovery of many nuclear proteins.

Recovery time-series after increasing heat-stress exposures in Caenorhabditis elegans measured by microarray

Project description:Increased exposure to heat-stress leads to an increasingly altered transcriptome in Caenorhabditis elegans. It is however unclear how the recovery from heat-stress progresses after increased heat-stress exposures. Hence, we exposed populations of the N2 strain to a 2, 3, 4, or 6 hour heat-shock of 35 degrees Celsius and took samples from 1 - 4 hours after termination of stress. This experiment was conduced in three biological replicates.
