Real-time quantitative PCR analysis of human embryonic stem cells, human neural stem cells and human foreskin fibroblast.

ABSTRACT: To address the question of whether v-myc modifies the stemness and potency of hNSCs, we first studied the expression of 90 genes related to stemness, self-renewal and differentiation in several v-myc immortalized cell lines (hNS1, hVM1, hCTX), non-immortalized neurospheres (hNPC derived from 9.5 and 10 weeks-old fetuses), hESCs (H9 and HS181) and human foreskin fibroblasts (hFF-1).

Transcription profiling of human neural progenitor cells (hNPC) derived from the developing cortex (CTX) and ventral midbrain (VM)

Project description:Neural progenitor cells (hNPC) derived from the developing human brain can be expanded in culture and subsequently differentiated into neurons and glia. They provide an interesting source of tissue for both modeling brain development and future cellular replacement therapies. It is becoming clear that hNPC are regionally and temporally specified depending on which brain region they were isolated from and its developmental stage. We show here that hNPC derived from the developing cortex (hNPCCTX) and ventral midbrain (hNPCVM) have similar morphological characteristics and express the progenitor cell marker nestin. However, hNPCCTX cultures were highly proliferative and produced large numbers of neurons, while hNPCVM divided slowly and produced less neurons but more astrocytes. Microarray analysis revealed a similar expression pattern for some stemness markers between the two growing cultures, overlaid with a regionally specific profile that identified some important differentially expressed neurogenic transcription factors. By over expressing one of these, the transcription factor ASCL1, we were able to regain neurogenesis from hNPCVM cultures which produced larger neurons with more neurites than hNPCCTX, but no fully mature dopamine neurons. Thus hNPC are regionally specified and can be induced to undergo neurogenesis following genetic manipulation. While this restores neuronal production with a region specific phenotype, it does not restore full neurochemical maturation which may require additional factors. Gene expression data from human neural stem cells (hNSCs) treated with 10 uM TCQA for 24 hours

Project description:TCQA promoted differentiation of hNSCs at least toward the neuronal lineage and suggests the possibility of using TCQA to promote neurogenesis. When differentiation is induced by growth factor withdrawal, decreased expression of stemness gene and increased expression of NSC fate-promoting genes can be observed. Therefore, we evaluated the effect of TCQA on global gene expression in hNSCs during differentiation at 24h to elucidate the neurogenesis-promoting effects of TCQA.

Transcriptomic profiles of human foreskin fibroblast cells in response to orf virus

Project description:Using RNA sequencing technology, we compared speciﬁc changes in the transcriptomic proﬁles in human foreskin fibroblast cells (HFF-1) following ORFV infection. ORFV speciﬁcally upregulated and downregulated a variety of genes, including genes involved in antiviral immune response, apoptosis, cell cycle and a series of signaling pathways, such as the IFN and p53-signaling pathways. A range of genes were downregulated at early stage. In contrast, the virus up-regulates a number of cellular genes at late stage.

p53 regulates viral gene expression during infection with human cytomegalovirus

Project description:Human cytomegalovirus (HCMV) is a common cause of morbidity and mortality in immunocompromised and immunosuppressed individuals. During infection, HCMV is known to employ host transcription factors to facilitate viral gene expression. To further understand the previously observed delayed viral replication and protein expression in p53 knockout cells, we conducted microarray analyses on p53+/+ and p53-/- immortalized fibroblast cell lines. At 12 hpi, applying the significance criteria defined above, no genes showed a significant difference in any of the comparisons. At 24 hpi, we found 19 genes to be significantly different in the THF versus p53-/- comparison and 14 genes to be significantly different in the HFF versus p53-/- comparison. Eleven genes were shared between HFF and THF cells when compared to p53-/- cells (UL30, UL54, UL63, UL65, UL69, UL75, UL110, UL122, IRL4, IRS1, TRS1, shown in bold in Table 1). Eight genes were unique in the THF versus p53-/- comparison (UL48, UL68, UL84, UL100, UL106, UL109, UL111, RL5A), and three genes in the HFF versus p53-/- cell comparison (UL14, UL16, US2). When comparing HFF and THF cells, only US2 showed a significant difference. The ratio (HFF versus p53-/- divided by THF versus p53-/-) of the expression levels of the eleven shared genes from the HFF versus p53-/- and THF versus p53-/- comparisons was an averaged 1.2, indicating that, with the exception of US2, these two cell lines behaved similarly with respect to p53-driven viral gene expression.
