Circulating microRNAs as non-invasive biomarkers for early detection of non-small-cell lung cancer

ABSTRACT: Plasma samples from 100 early stage (I to IIIA) non–small-cell lung cancer (NSCLC) patients and 100 non-cancer controls were screened for 754 circulating microRNAs via qRT-PCR, using TaqMan MicroRNA Arrays. Our objective was to identify a panel of circulating microRNAs in plasma that will contribute to early detection of lung cancer.

Project description:Exosomes were purified from 250 ul serum using ExoQuickTm. The presence of particles consistent in size with exosomes (60-150nm) was confirmed using a Nanosight LM10. miRNA was extracted from exosomes using an miRNeasy Serum/Plasma kit (Qiagen, #217184). miRNA was reversed transcribed using a TaqMan® microRNA Reverse Transcription Kit (Life technologies, #4366596). miRNA profiling was performed with a high throughput TaqMan® OpenArray® Human microRNA panel (Life technologies, #4461104). The panel consisted of probes for 754 human miRNAs that are based on miRNA sequences derived from Sanger miRBase v14. MegaplexTM Primer Human Pool A v2.1 and Human Pool B v2.0 or v3.0 The poor prognosis and rising incidence of oesophageal adenocarcinoma highlight the need for improved methods for detection of this cancer. Molecular biomarkers offer potential for this. The potential for circulating miRNAs as biomarkers in some other cancers has been shown, but circulating miRNAs have not been well characterized in oesophageal adenocarcinoma. This study investigated whether circulating miRNAs could be used to detect oesophageal adenocarcinoma.

Quantitative RT-PCR profiling of miRNA expression of non-small cell lung cancer patients against healthy controls

Project description:Lung cancer (LC) is one of the major cancers, with survival of patients dictated by the time of diagnosis. Cell-free circulating miRNAs have been proposed as candidate biomarkers for non-small cell lung cancer. The goal of this study was to profile the miRNAs in blood plasma of lung cancer patients diagnosed with squamous cell carcinoma (SCC, n=14) or adenocarcinoma (AD, n=6), and healthy individuals (HD) using miRCURY LNA miRNA qPCR Serum/Plasma Panel (Exiqon). Average Cq of detected assays was used for normalization of miRNA expression.

Differentially-expressed-transcriptome study of Non-Small Cell Lung Carcinoma

Project description:We systematically profiled the genome-wide alternative splicing events in Lung Adenocarcinoma (LUAD) and Lung Squomous Cell Carcinoma (LUSC) against Lung Control through Human Transcriptome Array2.0. Non-invasive Stage IIIA non-small cell lung cancer (NSCLC) is heterogeneous in nature which makes it difficult to predict, diagnose and prognose owing to lower 5-year survival rate and 75-85% brain or bone metastasis. Hence, we hypothesized to develop transcript-based signature to categorize Stage IIIA-NSCLC-LUAD and LUSC, as well as identify markers which could indicate towards prognosis of disease. We were able to molecularly-categorize LUAD- and LUSC-tissue more precisely through HTA array2.0.

The microRNA profile in neosquamous oesophageal mucosa following ablation of Barrett's oesophagus

Project description:mucosal biopsies were taken from patients at pre and post-argon plasma coagulation (APC) ablation of Barrett's oesophagus, and from healthy controls. Total RNA was extracted using Trizol. miRNA was reversed transcribed using a TaqMan® microRNA Reverse Transcription Kit (Life technologies, #4366596). miRNA profiling was performed with a high throughput TaqMan® OpenArray® Human microRNA panel (Life technologies, #4461104). The panel consisted of probes for 754 human miRNAs that are based on miRNA sequences derived from Sanger miRBase v14. Endoscopic surveillance remains the mainstay of cancer prevention in individuals with Barrett’s oesophagus, and definitive management by surgery or endoscopy is reserved for high grade dysplasia or cancer. There are several endoscopic treatments that are widely used for Barrett’s oesophagus, including argon plasma coagulation (APC). The normality of the regenerated neosquamous epithelium after ablation has been called into question. In the current study we profiled global microRNA expression in oesophageal mucosa before and after ablation of Barrett’s oesophagus using Argon plasma coagulation. Our aim was to investigate differences in microRNA expression between neosquamous and normal squamous tissues.

Project description:TaqMan low density array (TLDA) was carried out to screen of the profiles of circulating miRNAs in pooled plasma samples from healthy controls and pre-operative osteosarcoma patients. The expression changes of circulating miRNAs in osteosarcoma patients were identified. To select candidate plasma miRNAs for osteosarcoma detection and monitoring, we employed TLDA technique to screen expression levels of 739 miRNAs in pooled plasma samples from healthy controls and pre-operative osteosarcoma patients (each pooled from 10 individuals).
