Project description:Adoptive natural killer (NK) cell therapy in combination with hematopoietic stem cell transplantation (HSCT) can provide cure for acute myeloid leukemia (AML) but is in part dependent on variable NK cell reactivity. Adaptive NK cells are a highly potent NK cell subsets that can be utilized to maximize ‘missing-self’ reactivity against tumor cells. Project description:Natural Killer cells (NK), a major constituent of innate immune system, have the ability to kill the transformed and infected cells without prior sensitization; can be put to immunotherapeutic use against various malignancies. NK cells discriminate between normal cells and transformed cells via a balance of inhibitory and activating signals induced by interactions between NK cell receptors and target cell ligands. Present study investigates whether expansion of NK cells could augment their anti-myeloma (MM) activity. For NK cell expansion, peripheral blood mononuclear cells from healthy donors and myeloma patients were co-cultured with irradiated K562 cells transfected with 4-1BBL and membrane-bound IL15 (K562-mb15-41BBL). A genome-wide profiling approach was performed to identify gene expression signature in expanded NK (ENK) cells and non-expanded NK cells isolated from healthy donors and myeloma patients. A specific set of genes involved in proliferation, migration, adhesion, cytotoxicity, and activation were up regulated post expansion, also confirmed by flow cytometry. Exp-NK cells killed both allogeneic and autologous primary MM cells more avidly than non-exp-NK cells in vitro. Multiple receptors, particularly NKG2D, natural cytotoxicity receptors, and DNAM-1 contributed to target lysis, via a perforin mediated mechanism. In summary, vigorous expansion and high anti-MM activity both in vitro and in vivo provide the rationale for testing exp-NK cells in a clinical trial for high risk MM. Project description:Checkpoint blockage has revolutionized cancer treatment. NKG2A is an inhibitory receptor expressed by cytotoxic lymphocytes, including NK cells. In contrast to other checkpoint inhibitory antibodies, anti-NKG2A antibodies have shown only limited success. Here, we designed a Cas9-based strategy to delete KLRC1 from human NK cells. Electroporation of KLRC1-targeting Cas9-RNP efficiently eliminated NKG2A expression from primary human NK cells. NKG2A-deficient NK cells showed normal proliferation, only minor transcriptional changes related to enhanced NK cell activation and maintained their phenotype and licensing status. Genetic deletion of NKG2A fully bypassed HLA-E inhibition and further enhanced NK cell activity against various tumor cell lines, thereby outperforming anti-NKG2A antibodies. In combination with antibody-coating of tumor cells to induce antibody-dependent cellular cytotoxicity, genetic deletion of NKG2A independently promoted cytotoxicity. Thus, Cas9-mediated targeting of NKG2A is an effective way to target this important inhibitory checkpoint. This technique is easily amenable to adoptive cell therapy in the clinical setting, where NKG2A deletion will promote anti-tumor responses and may help NK cells to better infiltrate and persist in an inhibitory tumor microenvironment.

Project description:γδ-T lymphocytes are cells of the innate immune system exerting potent natural cytotoxicity against bacteria, viruses and tumours. This ability, coupled with their negligible alloreactivity, makes them attractive for adoptive immunotherapy approaches. To achieve a cell product suitable for clinical use, a protocol offering the possibility to generate and expand high numbers of polyclonal γδ-T cells is required. In this study, we designed a robust protocol to either manually or automatically expand high numbers of functional polyclonal memory γδ-T cells under good manufacturing practice conditions, with the additional possibility of gene-modification to improve anti-tumour activity, proving the feasibility of establishing an allogeneic “third-party T-cell bank” for off-the-shelf use. Engineered artificial antigen-presenting cells (aAPCs) expressing CD86/41BBL/CD40L and the cytomegalovirus (CMV)-antigen-pp65 were used for γδ-T cell production. The presence of CMV-pp65 and CD40L proved to be crucial for expansion of the memory Vδ1 subpopulation. To allow clinical translation and guarantee patient safety, aAPCs were stably transduced with an inducible suicide gene. The proposed cell product represents an attractive therapeutic option endowed with broad clinical applications, including treatment of viral infections in highly immunocompromised patients, treatment of aggressive malignancies refractory to conventional approaches and, ultimately, innovative platform for development of off-the-shelf CAR-T-cell products.

Project description:The harnessing of natural killer cells (NK cells) as adoptive anti-cancer therapy relies on sufficient numbers of functional NK cells. The use of autologous peripheral blood mononuclear cells (PBMCs) from respective allogeneic donors to boost NK cell proliferation ex vivo, has shown to be a suitable strategy to obtain high NK cell numbers for clinical applications. However, the mechanisms underlying the effect of PBMCs on NK cell proliferation have not been completely characterized. Thus, in this study, we performed a thorough analysis of the influence of autologous PBMCs on NK cell proliferation. We used irradiated autologous CD56-negative PBMCs stimulated with OKT3 that induced significantly increased expansion of purified CD56+CD3- NK cells, already after 5 days of culture (3.2 mean fold, n= 9) compared to standard cultures with only IL-2 supplement (1.3 mean fold n=90 proliferation, as observed when NK cells were cultured separately from the PBMCs through a permeable membrane. Depletion of B cells had no influence on the enhancement of NK cell proliferation, observing no changes in terms of fold expansion or cytokine profile, exemplified by high levels of TNF-α, IFN-γ, IL-6 and GM-CSF. In contrast the absence of T cells or monocytes reduced dramatically the proliferation of NK cells and abrogated cytokine production. Moreover, we analyzed differences in gene expression between proliferating and non-proliferating NK cells within this co-culture set up, to sought to determine changes in signalling pathways or phenotype related to a high proliferative status. The transcriptome analysis indicated that actively proliferating NK cells developed a differential activation response towards the stimuli provided during the co-culture compared to non-proliferating NK cells. In particular, differences in phenotype, expression of mitogen-activated protein kinases and zinc finger transcription factors could be demonstrated. In summary, our findings give rise to improve ex vivo expansion protocols of NK cells for future clinical applications.

Project description:Hhex encodes a homeobox transcriptional regulator important for embryonic development and hematopoiesis. Hhex is highly expressed in NK cells and its germline deletion results in significant defects in lymphoid development, including NK cells. To determine if Hhex is intrinsically required throughout NK cell development or for NK cell function, we generated mice that specifically lack Hhex in NK cells. NK cell numbers was dramatically reduced, while NK cell differentiation, IL-15 response and function at the cellular level remained largely normal in the absence of Hhex. Increased IL-15 availability failed to fully reverse NK lymphopenia following conditional Hhex deletion, suggesting that Hhex regulates developmental pathways extrinsic to those dependent on IL-15. Gene expression and functional genetic approaches revealed that Hhex regulates NK cell survival by repressing expression of the key apoptotic mediator, BIM. These data implicates Hhex as a novel transcription factor essential for NK cell homeostasis and immunity.
