Total RNA samples were preparated from Tibialis-anterior (TA) muscle and hearts of three P9 MG56-knockout mice and the corresponding wild-type mice were mixed and then subjected to Affymetrix Mus musculus microarrays.

Systematic analysis of cardiac transcriptome and proteome in Tmem43 knockout mice reveals the defect of heart muscle contraction

Project description:Here, we profiled the transcriptome and proteome of the hearts of the Tmem43 null mouse via RNA-Seq and mass spectral. Functional enrichments of the differentially expressed genes and differentially expressed proteins both revealed that the muscle contraction was the major pathway influenced by TMEM43 deficiency. Further study indicated that BMP2 signaling pathway and mitochondria may mediate TMEM43’s role in hearts. These results expand our knowledge of the roles of TMEM43 in hearts.

Proteomics analysis of tibialis anterior muscle and descending thoracic aorta from Mustn1 whole-body knockout mice and wild-type littermates

Project description:Skeletal muscle is a highly adaptive tissue that changes with many physiological stimuli and is composed of many cell types. Upon muscle injury, the concerted action of these cells is required to proceed through the process of inflammation, extracellular matrix remodeling, and restoration of function. To uncover novel genes and molecular pathways important for skeletal muscle remodeling and regeneration, we used a mouse hindlimb unloading and reloading protocol, and performed transcriptomics analysis. This study focuses on the microprotein Mustn1 (Musculoskeletal embryonic nuclear protein 1, also known as Mustang), whose gene expression is increased in muscle at the onset of hindlimb reloading, exercise, and injury. We generated a whole-body Mustn1 knockout mouse model and performed proteomics analysis of muscle and aorta.

Analysis of skeletal muscle gene expression upon mouse hindlimb unloading and reloading

Project description:To investigate the molecular mechanisms governing the transition of skeletal muscle from atrophy to compensatory regeneration and hypertrophy, we employed a mouse model involving hindlimb unloading and subsequent reloading, conducting a comprehensive analysis of global gene expression using RNA-sequencing (RNA-seq). Gastrocnemius muscle samples were obtained from three groups: control mice, mice subjected to 10 days of hindlimb unloading-induced muscle atrophy, and mice reintroduced to normal cage activity for 1 day following the unloading period (reloading).

Transcription profiling of mouse hearts from mutants lacking estrogen-related receptor alpha (ERRalpha), a nuclear receptor

Project description:Total RNA was isolated from 3 WT and 3 ERRalpha null hearts and independent hybridizations were performed using MOE430 2.0 microarrays. Expression profiling was conducted to determine changes in gene expression in hearts lacking ERRa. The expression of genes involved in heart and muscle development, muscle contraction, lipid metabolism, OxPhos, protein metabolism and transcription were affected by the loss of ERRa. Experiment Overall Design: 3 hearts from WT and 3 hearts from ERRalpha-null mice were used in the study. The expression of genes in the ERRalpha KO hearts were compared to the reference WT hearts.

Gene expression analysis in skeletal muscle and heart of 11 days-old TK2 knockout mice

Project description:TK2 deficiency causes severe mtDNA depeltion in several tissues, including skeletal muscle and heart. TK2 knockout mice grow slower and their skeletal muscles appeared significantly underdeveloped, whereas heart was close to normal size. We used microarrays in order to compare the transcriptomes in skeletal muscle and heart tissue of 11 days-old TK2 knockout pups with the sames tissues of wild-type pups at the same age. We collected skeletal muscle from the hind limb and hearts of three 11 days-old TK2 knockout and three wild-type pups and extracted total RNA. These RNA samples were used for hybridization in Affymetrix arrays.

Effects of spaceflight on murine skeletal muscle gene expression

Project description:Spaceflight results in a number of adaptations to skeletal muscle, including atrophy and shifts towards faster muscle fiber types. To identify changes in gene expression that may underlie these adaptations, microarray expression analysis was performed on gastrocnemius from mice flown on the STS-108 shuttle flight (11 days, 19 hours) versus mice maintained on earth for the same period. Additionally, to identify changes that were due to unloading and reloading, microarray analyses were conducted on calf muscle from ground-based mice subjected to hindlimb suspension (12 days) and mice subjected to hindlimb suspension plus a brief period of reloading (3.5 hours) to simulate the time between landing and sacrifice of the spaceflight mice.
