Knockdown of TSPAN8 gene expression in the SH-SY5Y neuroblastoma cell line

ABSTRACT: Bipolar disorder (BD) has an estimated heritability of about 80%. Different pathways and candidate genes may contribute to the pathogenesis of BD, but definite mechanisms are yet unresolved. In a previous study, we identified the single nucleotide polymorphism (SNP) rs4500567, located in the upstream region of Tetraspanin 8 (TSPAN8), to be associated with bipolar disorder (BD). The SNP´s minor allele results in a predicted binding site for the transcription factor Nuclear Factor Interleukin 3, regulated (NFIL3); luciferase assays confirmed allele-specific TSPAN8 expression levels.

Comparative genome analysis of exosomal mRNA derived from Tspan8-overexpressing and Tspan8-negative rat pancreas tumors

Project description:We recently described that tumor-derived exosomes containing the tetraspanin Tspan8 induce angiogenesis. However, not much is known about exosome-endothelial cell interactions and there are no clues on the role of tetraspanins, which are a constitutive component of exosomes. We used the stably Tspan8 expressing rat adenocarcinoma cell line AS-Tspan8, to explore how exosomal Tspan8 supports angiogenesis. Tspan8 contributed to a selective recruitment of proteins and mRNA into exosomes. A genome-wide microarray analysis revealed that both, AS- and AS-Tspan8-exosomes contain about 1500 transcripts, from which 285 are over 3 fold enriched in AS-Tspan8-exosomes. We confirmed high levels mRNA of transcription factors MycN and Spi-C, elongation factor B(SIII), TCEB-1, fibroblast growth factor binding protein 1 (FGFBP1) and growth and differentiation factor-3 (GDF3) in AS-Tspan8-exosomes compared to AS-exosomes and to the parental cells by RT-PCR. VEGF mRNA was not enriched in AS-Tspan8 cells or exosomes. Expression of CCL20, prothrombin, bone morphogenetic protein-2, several Ras and MAPK family members and a phosphatase2A subunit was below the detection level in exosomes.

Transcriptome profiling of the human dorsal striatum in bipolar disorder

Project description:Bipolar disorder (BD) is a highly heritable and heterogeneous mental illness whose manifestations often include impulsive and risk-taking behavior. This particular phenotype suggests that abnormal striatal function could be involved in BD etiology, yet most transcriptomic studies of this disorder have concentrated on cortical brain regions. We report the first transcriptome profiling by RNA-Seq of the human dorsal striatum comparing bipolar and control subjects. Differential expression analysis and functional pathway enrichment analysis were performed to identify changes in gene expression that correlate with BD status. Further co-expression and enrichment analyses were performed to identify sets of correlated genes that show association to BD. Total RNA samples were isolated from 36 postmortem dorsal striatum subjects (18 bipolar and 18 control) and sequenced. One outlier sample was removed and 35 samples (18 bipolar and 17 control) were analyzed.

We revealed neuronal cell-type-specific, pathophysiology-related DNA methylation changes in the prefrontal cortex (PFC) of BD patients, highlighting the importance of the neural epigenome for understanding BD.
