Project description:This study was undertaken to compare efficacy of MJC13 and the classic AR antagonist flutamide. Both compounds modulate endogenous AR target gene expression in Prostate cancer cells in a similar fashion despite different mechanisms of action. LNCaP cells were seeded within 6-well dishes at density of 1 à 100,000 within RPMI 1640 media supplemented with L-glutamine and 10% fetal bovine serum (FBS), day 2 the media replaced with RPMI 1640 media supplemented with L-glutamine and 10% charcoal-stripped FBS, day 3 the cells were treated with DMSO (control), 10nM DHT, 1μM Flutamide, 1μM Flutamide + 10nM DHT, 30μM MJC13, or 30μM MJC13 + 10nM DHT for 24 hours, after which total RNA was isolated.

Project description:2-Isopropylthioxanthone (2-ITX), a photoinitiator used in ink for printing of food packaging materials, has been detected in products like milk and fruit beverages. Previous in vitro research demonstrated 2-ITX to 1) exert agonistic effects on the aryl hydrocarbon receptor (AhR), 2) exert antagonistic effects on both the androgen (AR) and estrogen receptor (ER), and 3) to increase aromatase activity. To validate these findings in vivo, male rats were treated by gavage with 50, 150 or 500 mg 2-ITX/kg b.w. per day from postnatal day 23 to 53. For comparison, other groups were treated with 50 ng TCDD/kg b.w./day (AhR agonist), 30 mg bicalutamide/kg b.w./day (Casodex, AR antagonist) or 50 mg flutamide/kg b.w./day (AhR agonist and AR antagonist). 2-ITX, from the lowest dose onwards, decreased the weight of the ventral prostate, and from the middle dose onwards the weight of the seminal vesicles and coagulating gland. The highest dose of 2-ITX reduced the number of spermatozoa in the cauda of the epididymis. Similar effects, but more severe, were induced by flutamide and bicalutamide. Epididymal weight, caput sperm count and preputial separation were negatively affected by Casodex and flutamide but not by 2-ITX. The two highest doses of 2-ITX as well as TCDD increased liver weight. Whole genome mRNA expression profiling of the liver revealed similarity between the effects of 2-ITX and flutamide. In addition, many genes induced by TCDD were also induced by 2-ITX and flutamide. These results demonstrate 2-ITX to express AhR agonistic and AR antagonistic activities, both in vitro and in vivo.

Project description:Trenbolone and flutamide are prototypical model compounds for respectively androgen and anti-androgen modes of action. Trenbolone is an anabolic steroid used in cattle industry to increase weight gain and feed efficiency, and flutamide is a pharmaceutical used to treat prostate cancer. Androgens exert profound effects on the organization, development, and function of the male reproductive system through activation of androgen receptors (ARs). Flutamide, as an antiandrogen, was expected to temper the effects of trenbolone on androgen receptor (AR)-mediated gene expression. Female fathead minnows (Pimephales promelas) were exposed via the water to 0.05, 0.5 or 5 µg 17β-trenbolone/L; 50, 150 or 500 µg flutamide/L; or to a mixture of 0.5 μg/L trenbolone and 500 μg/L flutamide. After a 48 hr constant exposure gene expression in gonads was analyzed using a fathead minnow-specific 22,000-gene microarray. Trenbolone significantly changed the expression (P < 0.05) of roughly 750 different genes, while flutamide changed the expression of 1420 genes. Most of the genes that were regulated were distinct for the two chemicals. However, 70 genes were reciprocally regulated by the two treatments. Myelocytomatosis viral oncogene homolog (Myc), Yin Yang 1 (YY1) and interferon regulator factor 1 (IRF1) were among the important transcription factors reciprocally regulated by trenbolone and flutamide, suggesting they may be specifically regulated by the AR. These regulatory molecules, in conjunction with other reciprocally regulated transcription factors, may function as early molecular switches to control phenotypic changes in gonad tissue architecture and function.

Project description:Approved drugs are invaluable tools to study biochemical pathways and further characterization of these compounds may lead to repurposing of single drugs or combinations. Here, we describe a collection of 308 small molecules representing the diversity of structures and molecular targets of all FDA-approved chemical entities. The CeMM Library of Unique Drugs (CLOUD) covers prodrugs and active forms at pharmacologically relevant concentrations and is ideally suited for combinatorial studies. We screened pairwise combinations of CLOUD drugs for impairment of cancer cell viability and discovered the synergistic interaction between flutamide and phenprocoumon (PPC). The combination of these two drugs modulates the stability of the androgen receptor (AR) and resensitizes AR mutant prostate cancer cells to flutamide. Mechanistically, we show that the AR is a substrate for γ-carboxylation, a post-translational modification inhibited by PPC. Collectively, our data suggest that PPC might be repurposed to tackle resistance to antiandrogens in prostate cancer patients.

Project description:Aiming to identify insulin-independent modulators of glucose homeostasis, we performed a drug screen on zebrafish insulin (ins) mutants and identified androgen receptor (AR) antagonists. To investigate how AR antagonism mediates glucose level reduction in ins mutants, we evaluated the effects of antagonist treatment using transcriptomic studies. RNA-Seq analyses were performed on 120 hours post fertilization (hpf) ins mutants treated with Flutamide or Cyproterone starting at 84 hpf compared to vehicle (DMSO) treated mutants.

Project description:Inhibition of androgen-receptor (AR)signaling is the foundation of therapeutic regimens for advanced prostate cancers. However, nearly all patientsdevelop resistance and some never respond. To identify genes that mediate resistance to androgen ablation therapy, we performed an open reading frame (ORF) expression screen of17,255 ORFs and found that the transcription factorCREB5robustly conferred resistance to androgen deprivation and AR inhibition by enzalutamide. CREB5 overexpressionincreased enzalutamide-resistance 45-fold, enhanced resistance of tumor xenografts. CREB5 expression was also essential for enzalutamide-resistance patient-derived organoids. In clinical mCRPC, CREB5is frequently amplified and itsoverexpression positively correlated with castration resistance gene signatures including those of MYC and cell cycle. Mechanistically, CREB5 directly interacted at AR binding sites andenhanced site-specific AR bindingin enzalutamide. This dysregulated expression of 393 AR target genes including MYC and CDK1.These observations identifyCREB5asadriver of enzalutamide-resistance in a subset of advancedprostate cancer.

Project description:Mutations of the androgen receptor (AR) associated with prostate cancer and androgen insensitivity syndrome may profoundly influence its structure, protein interaction network and binding to chromatin, resulting in altered transcription signatures and drug responses. Current structural information fails to explain the effect of pathological mutations on AR structure-function relationship. Here we have thoroughly studied the effects of selected mutations that span the complete dimer interface of AR ligand-binding domain (AR-LBD) using X-ray crystallography in combination with in vitro, in silico, and cell-based assays. We show that these variants alter AR-dependent transcription and responses to anti-androgens by inducing a previously undescribed allosteric switch in the AR-LBD that increases exposure of residue Arg761 and ultimately leads to its enhanced methylation. We also corroborate the relevance of residues Arg761 and Tyr764 for AR dimerization and function. Together, our results reveal allosteric coupling of AR dimerization and post-translational modifications as a disease mechanism with implications for precision medicine.
