MiRNA Real-time quantitative PCR analysis of EAE CD4+T cells

ABSTRACT: EAE is a mouse model of human multiple sclerosis. We used miRNA low density array to screen abnormally expressed miRNAs in autoimmune CD4+T cells.Generally, splenic CD4+T cells were isolated from three groups: healthy C57BL/6 mice, C57BL/6 mice with the induction of EAE for 16 days, and C57BL/6 mice with the induction of EAE for 32 days. Then we did miRNA profilings on these samples.

We used miRNA low density array to screen abnormally expressed miRNAs in autoimmune CD4+T cells.Generally, splenic CD4+T cells were isolated from three groups: healthy C57BL/6 mice, C57BL/6 mice with the induction of EAE for 16 days, and C57BL/6 mice with the induction of EAE for 32 days. Then we did miRNA profilings on these samples. Splenic CD4+T cells were isolated from the above three groups. Each group consist of six mice. Equal amount total RNA from each mouse was pooled prior to miRNA expression analysis.

Gene expression profiling of mouse experimental autoimmune encephalomyelitis (EAE) from spinal cord preparations of opticospinal EAE (OSE) and MOG-induced EAE

Project description:Genetic opticospinal EAE (OSE) and MOG-induced EAE (MOG-EAE) are two experimental autoimmune encephalomyelitis (EAE) mouse models of human multiple sclerosis. For the OSE model, double-transgenic 2D2 (TCRMOG) x IgHMOG mice were used. For MOG-EAE, wildtype C57BL/6 mice were immunized with a MOG peptide consisting of the amino acids 35-55, administered in complete Freund’s adjuvant containing 5mg / ml Mycobacterium tuberculosi. The severity of EAE was rated on the scale 0: healthy animal; 1: animal with a flaccid tail; [...]; 4: animal with both hind legs paralyzed. The case groups in the experiment were: OSE1: OSE with disease score 1; OSE4: OSE with disease score 4; MOG4: MOG-EAE injected with both MOG and adjuvant, with disease score 4. The control groups in the experiment were: OSE0: OSE with disease score 0; CFA: C57BL/6 mice injected only with adjuvant (no MOG); WT: Wildtype C57BL/6 mice. The aim of the experiment was to assess gene expression differences 1) between OSE4 and OSE0, 2) between OSE1 and OSE0, and 3) between MOG4 and CFA. For control, WT was compared to OSE0 and CFA. Subsequently, differentially expressed transcripts were compared, first, between the OSE4 vs. OSE0 and the MOG4 vs. CFA contrasts (different EAE models) and, second, between the OSE4 vs. OSE0 and the OSE1 vs. OSE0 contrasts (different EAE severity).

Gene expression analysis of WT Th17 versus STAT4 deficient Th17 cells from EAE mice

Project description:The purpose of this study is to compare transcriptional profiles of WT and STAT4-deficient Th17 cultured cells from mice with experimental autoimmune encephalomyelitis (EAE) using RNA sequencing. WT and STAT4 deficient splenocytes from EAE immunized mice were cultured with MOG peptide under Th17 conditions for three days, and then total RNA was extracted from CD4 T cells for sequencing. Differential gene expression was determined using the DESeq2 algorithm. These data reveal a previously unrecognized role for STAT4 in Th17 gene expression and function.

We examined the mechanism by which NR4A2 can control the pathogencity of T cells in CNS autoimmune disease. Project description:The aim of the experiment is to characterize the genetics and mechanisms of the inflammatory response after induction of experimental autoimmune encephalomyelitis (EAE), a model of multiple sclerosis, by studying the genome wide expression in the spleen in late disease. A backcross was created between EAE-susceptible DA and EAE-resistant PVG rats. At day 35 after induction of EAE with myelin oligodendrocyte glycoprotein (MOG) in these rats, spleens were taken for transcriptional profiling.

Neuron-specific RNA responses to EAE and cuprizone

Project description:Ribotag mice were crossed to synapsin-CRE mice to induce expression of HA-tagged Rpl22 subunits in neurons. Mice were fed cuprizone to induce demyelination or were induced with MOG(35-55) peptide to establish experimental autoimmune encephalomyelitis. Brain was collected from EAE mice at 18 days after induction (peak disease) and from cuprizone-fed mice at 6 weeks on chow. Brain was also collected from unmanipulated control mice fed normal chow. Brain lysates were immunoprecipated with anti-HA antibody to isolated neuron-specific polyribosomes. RNA was extracted from the IP and analyzed by Affymetrix GeneChip® Mouse Gene 2.0 ST Arrays.

Transcriptome analysis of CD4+ T cells in experimental autoimmune encephalitomyelitis

Project description:In this study we determined whole genome gene expression of murine naive CD4+ T cells, in vitro differentiated Th17 cells, and CD4+ T cells isolated from experimental autoimmune encephalitomyelitis (EAE)-affected animals either after adoptive transfer of Th17 cells or after immunization with MOG35-55-peptide. The overall goal was to identify candidate genes involved in T cell pathology, encephalitogenicity and plasticity. These findings could then be correlated to multiple sclerosis pathology. Naive CD4+ T cells were isolated from B6.2d2 transgenic mice with MOG-specific T cell receptors and differentiated in vitro into Th17 cells. These Th17 cells were adoptively transferred into lymphopenic RAG1-/- mice to induce EAE. Further, EAE was induced by immunizing wild-type C57BL/6 mice with MOG35-55 peptide. RNA was extracted from naive CD4+ T cells, Th17 cells, and from CD4+ T cells isolated from the CNS of EAE-affected mice for gene expression analysis. Replicates from three independent experiments were analyzed.
