We performed a microarray analysis to get insights into the differentially regulated pathways between WT and KO under inflammatory conditions. Total RNA obtained from isolated peritoneal macrophages isolated from DUSP3-/- and WT mice 36 hours after challenging them in vivo with PBS or with 6mg/kg of LPS.

Cyclooxygenase gene (Ptgs) exchange and differentially regulated inflammatory pathways in peritoneal macrophages

Project description:The aim of the current study was to characterize the differential inflammatory signaling pathway of COX isoforms during systemic inflammation in mice. Peritoneal macrophages from mice challenged with either lipopolysaccharide (2 mg/kg body weight, WT, COX-2>COX-1, COX-1>COX-2, and Reversa mice) or PBS (Control, WT mice) were harvested. RNA (100 ng) was used to determine gene expression changes utilizing pre-built Mouse Inflammation V2 CodeSets to carry out Nanostring analysis. Our results indicate inflammatory networks can be maintained by isoform exchange in inflamed macrophages. However, COX-1>COX-2 macrophages show reduced activation of inflammatory signaling pathways, indicating that COX-1 may be replaced by COX-2 within this complex milieu, but not vice versa.

Project description:We compared the transcriptomes of mesenchymal cells and immune cells in mice sensitized and challenged with house dust mite (HDM) extracts and those in control PBS-treated mice, using single cell RNA-seq (scRNA-seq). Data with mesenchymal cells and data with immune cells were obtained from separate experiments. For scRNA-seq of mesenchymal cells, each sample was pooled from cells from 3 mice and 2 different samples per group (for a total of 6 mice) were used. Casp8- and Casp11-dependent gene expression in ileal mucosa of LPS-challenged male mice

Project description:The execution of shock following high dose E. coli lipopolysaccharide (LPS) or bacterial sepsis in mice required pro-apoptotic caspase-8 in addition to pro-pyroptotic caspase-11 and gasdermin d. Hematopoietic cells produced MyD88- and TRIF-dependent inflammatory cytokines sufficient to initiate LPS shock independent of caspase-8 or caspase-11. Both proteases had to be present to support tissue injury, dependent upon TNF and interferon , first observed in the small intestine and later in spleen and thymus. Caspase-11 enhanced the activation of caspase-8 and extrinsic cell death machinery within the lower small intestine. Neither caspase-8 nor caspase-11 was individually sufficient for shock, but collaborated to amplify the inflammatory signals associated with tissue injury. Therefore, combined pyroptotic and apoptotic signaling mediated endotoxemia independent of RIPK1 kinase activity and RIPK3 function. These observations bring to light the relevance of tissue compartmentalization during inflammatory signaling in vivo where cytokines execute diverse cell death pathways and tissue damage.
