Project description:Mice with thyroid-specific expression of oncogenic BRAF (Tg-Braf) develop papillary thyroid cancers (PTC) that are locally invasive and have well-defined foci of poorly differentiated thyroid carcinoma (PDTC). To investigate the PTC-PDTC progression, we performed a microarray analysis using RNA from paired samples of PDTC and PTC collected from the same animals by laser capture microdissection. Analysis of 8 paired samples revealed a profound deregulation of genes involved in cell adhesion and intracellular junctions, with changes consistent with an epithelial-mesenchymal transition (EMT). This was confirmed by IHC, as vimentin expression was increased and E-cadherin lost in PDTC compared to adjacent PTC. Moreover, PDTC stained positively for phospho-Smad2, suggesting a role for transforming growth factor-beta (TGFbeta) in mediating this process. Accordingly, TGFbeta induced EMT in primary cultures of thyroid cells from Tg-Braf mice, whereas wild-type thyroid cells retained their epithelial features. TGFbeta-induced Smad2 phosphorylation, transcriptional activity and induction of EMT required MAPK pathway activation in Tg-Braf thyrocytes. Hence, tumor initiation by oncogenic BRAF renders thyroid cells susceptible to TGFbeta-induced EMT, through a MAPK-dependent process. Comparing the transcription profiles of 8 pairs of murine poorly differentiated thyroid cancer and papillary thyroid cancer collected from the same animals by laser capture microdissection. Co-hybridizations were done in triplicate with a single dye flip.

Project description:We profiled the gene expression of 11 anaplastic thyroid carcinomas (ATC), 49 papillary thyroid carcinomas (PTC) and 45 normal thyroids (N) We hibridized a series of anaplastic thyroid carcinomas (ATC) and papillary thyroid carcinomas (PTC) onto Affymetrix U133 Plus 2.0 arrays. ATCs were obtained from different hospitals in France and Belgium. Paired RNA samples of PTCs and non-tumoral thyroid tissues were obtained from Ukraine via the Chernobyl Tissue Bank (www.chernobyltissuebank.com). Diagnoses were confirmed by the members of the International Pathology Panel of the Chernobyl Tissue Bank.

Project description:Papillary thyroid carcinoma (PTC) is the most common type of thyroid cancer. Histologically normal appearing tissue adjacent to the tumor (NAT) from PTC patients is commonly used as a control in thyroid cancer studies, but the molecular characteristics of NAT tissue are not well characterized. The aim of this study was to characterize the global gene expression profile of NAT and compare it with those of normal and tumor thyroid tissues. We performed RNA sequencing of three categories of thyroid tissue samples including NAT, normal thyroid (N), and PTC tumor (T). Transcriptome analysis shows that NAT presents a unique gene expression profile, which was not associated with sex and the presence of lymphocytic thyroiditis. Among the differentially expressed genes (DEGs) of NAT vs N, 247 coding genes and 6 noncoding genes have been reported as cancer genes, being involved in cell proliferation, apoptosis, and tumorigenesis. In addition, dysregulated expression of ribosomal and small RNAs was identified in NAT. IPA analyses revealed that “Cellular Response to Therapeutics, Cardiovascular Disease, Organismal Injury and Abnormalities, Cancer, and Cellular Movement” appeared as major dysregulated pathways in the NAT tissues. This study provides a comprehensive study of the PTC transcriptome, which provides improved insight into the complexity of gene expression changes during PTC tumorigenesis.

We used microarrays to identify gene signature of radiation-induced papillary thyroid carcinomas To identify molecular differences between radiation-induced (Exposed to Chernobyl Radiation, ECR) and sporadic PTC, we investigated 65 childhood/young adult PTC samples using DNA microarray (Affymetrix, Human Genome U133 2.0 Plus). The PTC samples were from patients born either before (33 ECR cases) or at least 9 months after (32 non-ECR cases) the Chernobyl catastrophe. Multofactoral analyses were performed in order to define some additional factors that could have impact on the gene expression profile. Morover the microarray data were validated with the QPCR reaction and exon arrays.
