Lung metastasis 100% vs 0%

ABSTRACT: Lung metastatic potential of the 29 HNSCC was examined with tail vein metastatic assay as described previously. 1 ×10E6 HNSCC cells in a volume of 200 µL were injected into the lateral tail vein using a 30 gauge needle. Eight to 10 mice were prepared for each cell line. Mice were euthanized using carbon dioxide asphyxiation when they lost more than 15% of their pre-injection body weight or at 90 days after cell injection. Necropsy was performed, and lung was harvested. The lungs were then evaluated for the presence of lung metastases and also weighed.

The MS data for Walker-256 lung metastasis rat model by Orbitrap Fusion Lumos

Project description:We performed a lung metastasis of a rat model was established by tail-vein injection of Walker-256 cells. Urine samples were collected at days 2, 4, 6 and 9 after injection, and the urinary proteomes were profiled. On days 2, 4, 6 and 9, 11, 25, 34 and 44 differential proteins were identified in 7 lung metastatic rats. Seventeen and 18 differential proteins were identified on the second day and the fourth day respectively, and all these 17 and 18 differential proteins had been reported to associate with both lung cancer and breast cancer. A total of 9 differential proteins were identified in all 7 lung metastatic rats at more than one time point but none of them were identified in the subcutaneous rat model. Our results indicate that (1) the urine proteome changed significantly, even on the second day after tail-vein injection of Walker-256 cells and that (2) the urinary differential proteins were different in Walker-256 lung metastatic and subcutaneous models.

Comparative gene expression profiling analysis of RNA-seq data for mCherryHigh and DTC populations compared to mCherryLow metastatic populations [mCherry Pops and DTCs]

Project description:To investigate characteristics of the Gstt1high metastatic subppulation in pancreatic cancer, lung metastatic cell line was generated to express mCherry from the Gstt1 endogenous promoter. Mice were injected via tail vein with mixed populations until evidence of metastatic disease, or 5 days after injection for DTC sorting. Upon evidence of metastatic disease, mice were euthanized and lung tissues were digested followed by sorting of mCherry tumor cell populations (top 15-20%) gated on GFP. DTCs were collected 5 days after injection by gating and sorting on GFP. We then performed gene expression profiling analysis using data obtained from RNA-seq comparing mCherryHigh and low populations.

Project description:A highly metastatic breast cancer cell line, 4T1, was used to generate stable Wnt5a expressing and vector only control cells. Cells were generated using lentivirus infection and selection with blasticidin. Expression of Wnt5a was confirmed using western blot. Cell behaviour was characterized. Wnt5a expressing cells exhibited reduced migration in a transwell assay and reduced metastasis in a tail vein injection assay. Growth was not significantly affected.

Project description:Cancer staging and treatment frequently assume a binary division of tumors into localized or metastatic cancers. We proposed a state of metastatic disease defined by the number of metastases termed oligometastases. Patients with oligometastatic disease may be cured with localized methods of cancer treatment. We analyzed miRNA expression from paraffin blocks of primary or metastatic tumor samples derived from oligometastatic (? 5 metastases) patients treated with high dose localized radiotherapy. We report patterns of miRNA expression in the metastatic and primary tumor samples that identify patients who failed to progress to widespread polymetastases. We created a model of oligometastases of human tumors in immune compromised mice. The miRNA patterns of gene expression derived from patients accurately identified oligometastatic patterns in the mouse model as compared to animals that developed widespread metastases. MiRNA signatures may identify patients most likely to benefit from aggressive curative treatment of limited metastatic disease. Injection of MDA-MB-435-GFP cancer cells into the mammary fat pad of female athymic mice to develop spontaneous macroscopic lung metastasis. Tail vein experimental lung colonization assay was performed to model the development of MDA-MB-435-GFP Oligo- or Poly-metastases in the lung in vivo. Cell lines: Total RNA were derived from MDA-MB-435-L1-GFP (Ol-like) or MDA-MB-435-L1Mic (Poly-like) cell lines.
