B Cells Polarize Pathogenic Inflammatory T Helper Subsets through ICOSL-Dependent Glycolysis

Project description:B cells constitute abundant cellular components in inflamed human tissues, but their role in pathogenesis of inflammatory T helper (Th) subsets is still unclear. Here, we demonstrate that B cells, particularly resting naïve B cells, have a previously unrecognized helper function that is involved in shaping the metabolic process and subsequent inflammatory differentiation of T-cell-receptor-primed Th cells. ICOS/ICOSL axis-mediated glucose incorporation and utilization were crucial for inflammatory Th subset induction by B cells, and activation of mTOR was critical for T cell glycolysis in this process. Consistently, upon encountering ICOSL+ B cells, activated effector memory Th cells from patients with rheumatoid arthritis or systemic lupus erythematosus spontaneously differentiated into inflammatory Th subsets. Immunotherapy using an anti-CD20 antibody that specifically depleted B cells in patients with rheumatoid arthritis efficiently abrogated the capabilities of memory Th cells to incorporate and utilize glucose, thereby impairing the pathogenic differentiation of inflammatory Th subsets. To examine the metabolic process of Th cells after interacting with autologous B cells, we applied RNA-sequencing to analyze their transcriptional profiles.

RNA-seq of human CD4+ T cell subsets

Project description:After activation, CD4+ T helper (Th) cells differentiate into functionally specialized populations that coordinate distinct immune responses and protect against different types of pathogens. In humans, these effector and memory Th cell subsets can be readily identified in peripheral blood based on their differential expression of chemokine receptors that govern their homeostatic and inflammatory trafficking. Foxp3+ regulatory T (Treg) cells can also be divided into subsets that phenotypically mirror each of these effector populations, and share expression of key transcription factors and effector cytokines. In this study, we performed comprehensive transcriptional profiling of 11 phenotypically distinct Th and Treg cell subsets sorted from peripheral blood of healthy individuals. Despite their shared phenotypes, we found that mirror Th and Treg subsets were transcriptionally dissimilar, and that Treg cell populations showed limited transcriptional diversity compared to Th cells. We identified core transcriptional signatures shared across all Th and Treg cell populations, and unique signatures that define each of the Th or Treg populations. Finally, we applied these signatures to bulk Th and Treg RNA-seq data and found enrichment of specific Th and Treg cell populations in different human tissues. These results further define the molecular basis for the functional specialization and differentiation of Th and Treg cell populations, and provide a new resource for examining Th and Treg specialization in RNA-seq data.

Immunomodulation and T helper TH?/TH? response polarization by CeO? and TiO? nanoparticles.

Project description:Immunomodulation by nanoparticles, especially as related to the biochemical properties of these unique materials, has scarcely been explored. In an in vitro model of human immunity, we demonstrate two catalytic nanoparticles, TiO? (oxidant) and CeO? (antioxidant), have nearly opposite effects on human dendritic cells and T helper (T(H)) cells. For example, whereas TiO? nanoparticles potentiated DC maturation that led towards T(H)1-biased responses, treatment with antioxidant CeO? nanoparticles induced APCs to secrete the anti-inflammatory cytokine, IL-10, and induce a T(H)2-dominated T cell profile. In subsequent studies, we demonstrate these results are likely explained by the disparate capacities of the nanoparticles to modulate ROS, since TiO?, but not CeO? NPs, induced inflammatory responses through an ROS/inflammasome/IL-1? pathway. This novel capacity of metallic NPs to regulate innate and adaptive immunity in profoundly different directions via their ability to modulate dendritic cell function has strong implications for human health since unintentional exposure to these materials is common in modern societies.
