Project description:The purpose of this study was to define, at the single cell level, the transcriptionnal profile of murine tail epidermal basal cells during potsnatal growth and to compare with adult homeostatic basal cells. The analysis was performed on Lgr5DTR-EGFP mice (Tian et al., 2011)(knockin mice containing an Enhanced Green Fluorescent Protein (EGFP) under the control of the Lgr5 regulatory region), allowing to identify and exclude Lgr5-expressing cells of the bulge and basal cells of the interfollicular epidermis were enriched using EGFP negative, CD34 negative, alpha6 integrin positive gating. Our single-cell RNA sequencing revealed that epidermal basal cells at P7 form a more uniform population compared to adult basal cells and that heterogeneity starts to appear around P30.

Streamlined Single-cell Proteomics by an Integrated Microfluidic Chip and Data-independent Acquisition Mass Spectrometry

Project description:Single-cell proteomics can reveal cellular phenotypic heterogeneity and cell-specific functional networks underlying biological processes. Here, we present a streamlined workflow combining microfluidic chips for all-in-one proteomic sample preparation and data-independent acquisition (DIA) mass spectrometry (MS) for proteomics analysis down to the single-cell level. The proteomics chips enable multiplexed and automated cell isolation/counting/imaging and sample processing in a single device. Combining chip-based sample handling with DIA-MS using project-specific mass spectral libraries, we profile on average ~1,500 protein groups across 20 single mammalian cells. Applying the chip-DIA workflow to profile the proteomes of adherent and non-adherent malignant cells, we cover a dynamic range of 5 orders of magnitude with good reproducibility and <16% missing values between runs. Taken together, the chip-DIA workflow offers all-in-one cell characterization, analytical sensitivity and robustness, and the option to add additional functionalities in the future, thus providing a basis for advanced single-cell proteomics applications.

Single cell analysis of murine Dendritic Epidermal T Cells (DETC)

Project description:Purpose: To establish the steady state heterogeneity of murine Dendritic Epidermal T Cells (DETC) and to investigate the function of their constitutive interaction with neighbouring keratinocytes via Skint1 Methods: CD3+ cells were FACS purified from ear epidermal suspensions prepared from FVB mice administered isotype control or anti-Skint1 antibody 7d prior, then loaded into the 10x Genomics Chromium Platform and sequenced using Illumina HiSeq 4000. Conclusions: This analysis identifies four states of DETC in unperturbed epidermis, and reveals that DETC TCR sensing of keratinocytes via Skint1 potentially enables natural immune surveillance and maintains their thymic maturation signature in the periphery
