Identification of gene expression biomarkers in urine for the non-invasive detection of bladder cancer

ABSTRACT: Fifty patient urine samples diagnosed as high-grade urothelial carcinoma (HGUC) or benign were evaluated for bladder cancer via urine cytology. RNA was isolated and analyzed by microarray to identify a panel of biomarkers differentially expressed in HGUC and benign.

Project description:The management of bladder cancer faces multiple challenges concerning the diagnostics and fol-low-up approaches. The standard diagnostic test is invasive cystoscopy. Urine cytology and re-cently proposed urine-based biomarkers still could not replace cystoscopy, thus prompting calls for improvements. Here, we explore urine liquid biopsy to detect cancer mutations for therapeutic and prognostic assessment and subsequently evaluate the utility of urine as a suitable specimen in diagnosing bladder cancer. Our findings show that the analysis of pre-and post-operative urine enables the characterization of specific tumor mutations, which can be used to diagnose and evaluate a patientÃ¢Â€Â™s prognosis and therapy response.

Project description:Total RNAs were subjected to RNA isolation and RNA-seq analyses were generated by deep sequencing using Illumina sequencing platform. We performed RNA sequencing on three matched muscle-invasive bladder cancer (MIBC) tissues with superficial tumor regions and deep infiltrating tumor regions in order to assess its potential as biomarkers. Quantitative real-time PCR were used to validate the expression pattern of mRNAs and lncRNAs in urine exosomes. Bioinformatic tools including Gene ontology, and KEGG pathway analysis were utilized.

Molecular Biomarker Signature for Bladder Cancer Detection

Project description:In this study we applied differential gene expression analysis to exfoliated human urothelia obtained from patients of known bladder disease status. Selected targets from the microarray data were validated in an independent set of samples using a quantitative PCR approach. Total RNA was extracted from 52 samples of human urothelial cancer cells (C) and from 40 samples of human non-cancer (NC) urothelial cells. Fragmented, biotinylated cRNA was hybridized to Affymetrix Human Genome U133 Plus 2.0 microarrays.
