Definition of the AmrZ regulon under a twitching-promoting condition

Project description:A novel approach using cellophane sheets was designed to promote Pseudomonas aeruginosa twitching motility, as well as to provide convenience for cell harvest. The AmrZ regulon was defined under this specific growth condition by microarray analysis. WT P. aeruginosa and amrZ mutants were cultivated under this novel condition, and their transcriptional profiles were defined. WT P. aeruginosa strains include PAO1 and WFPA510 (complemented strain), while amrZ mutants include WFPA205 (ΔamrZ) and WFPA513 (AmrZV20A, DNA binding deficient). Each strain has 3 biological replicates.

Project description:We used the AHL-degrading enzyme AiiA-lactonase to interrogate the evolution of the LasR-dependent QS regulon of P. aeruginosa in conditions where QS is required to obtain carbon and energy, and show that populations of P. aeruginosa adaptively reduced the size of their QS regulon over ~1000 generations.

A Distinct QscR Regulon in the Pseudomonas aeruginosa Quorum Sensing Circuit

Project description:To better understand the role of QscR in P. aeruginosa gene regulation and to better understand the relationship between QscR, LasR and RhlR control of gene expression we used transcription profiling to identify a QscR-dependent regulon. Our analysis revealed that QscR activates some genes and represses others. Some of the repressed genes are not regulated by the LasR-I or RhlR-I systems while others are. The LasI-generated 3-oxododecanoyl-homoserine lactone serves as a signal molecule for QscR. Thus QscR appears to be an integral component of the P. aeruginosa quorum sensing circuitry. QscR uses the LasI-generated acyl-homoserine lactone signal and controls a specific regulon that overlaps with the already overlapping LasR and RhlR-dependent regulons. Using RNA Seq to define the regulon of the Pseudomonas aeruginosa transcription factor Anr in low-oxygen colony biofilms.

Project description:Purpose : The goal of this study was to use RNA Seq to define the regulon of the transciption factor Anr by comparing global transcriptional profiles of Pseudomonas aeruginosa strain PAO1 and a clinical isolate with their isogenic ∆anr mutants, grown in colony biofilms at 1% oxygen. Methods : mRNA profiles were generated for laboratory strain PAO1 and for a clinical isolate J215, as well as for ∆anr derivatives of each strain, in duplicate, by deep sequencing. Strains were grown for 12 hours in colony biofilms at 1% O2, 5% CO2 prior to RNA harvest. Ribosomal and transfer RNAs were removed using the MICROBExpress kit (Life Technologies). mRNA reads were trimmed and mapped to the PAO1 NC_002516 reference genome from NCBI using the ClC Genomics Workbench platform and defaut parameters.
