Project description:The expression signature of lncRNAs and mRNAs was analyzed in fibrotic tissue from peri-infarct region of mouse heart. We fund that fifty seven differentially expressed lncRNAs and 20 differentially expressed mRNAs are associated with 8 signaling pathways which are involved in the development of cardiac fibrosis, indicating a potential role of lncRNAs in cardiac fibrosis.

Project description:The challenge of oligohydramnios study is multiple causes of oligohydramnios. Long noncoding RNAs (lncRNAs) is a set of RNAs that has been proved to function in multiple biological process. Currently, little is know about their expression and possible role in oligohydramnios. Total RNA was isolated from fetal membranes ressected from oligohydramnios pregnant women (OR) and normal amniotic fluid control (Normal).RNA-sequencing (RNA-seq) obtain that a total of 801 lncRNAs and 367 mRNAs were differentially expressed in OR. Of which, 638 lncRNAs and 189 mRNAs were upregulated, and 163 lncRNAs and 178 mRNAs were downregulated. Of these lncRNAs, 566 of them were intergenic lncRNA, 351 were intronic antisense lncRNA，and 300 natural antisense. The differentially expressed lncRNA were primary located in chromosome 2, 1 and 11. KEGG enrichment pathways revealed the differential expressed mRNAs were enriched in pathway in cancer, Ras signaling pathway, TNF signaling pathway, focal adhesion, and chemokine signaling pathway. The qRT-PCR result confirmed that LINC00515 and RP11-388P9.2 were upregulated in OR. Furthermore, the constructed lncRNA-miRNA-mRNA regulatory network revealed TNR, CFTR, ABCC2, ABCA12, and COL9A2 as the candidate targets of LINC00515 and RP11-388P9.2. A wide range of lncRNAs were alert in OR, in particularly, LINC00515 and RP11-388P9.2 were confirmed to be uprgulated in OR, and their predicted downstream targets were transport and tissue growth and development associated. Further study focused on the role of differential expressed lncRNAs such as LINC00515 and RP11-388P9.2 would provide more insight into the pathophysiology in OR

Project description:A few reports have implicated specific lncRNAs in cardiac development or failure, but precise details of lncRNAs expressed in hearts and how their expression may be altered during embryonic heart development or by adult heart disease is unknown. By comparing lncRNA profiles of normal embryonic (~E14), normal adult, and hypertrophied adult hearts we defined a distinct fetal lncRNA abundance signature that includes 157 lncRNAs differentially expressed compared to adults (fold-change ≥ 50%, FDR=0.02), and which was only poorly recapitulated in hypertrophied hearts (17 differentially expressed lncRNAs; 13 of these observed in embryonic hearts). Analysis of protein-coding mRNAs from the same samples identified 22 concordantly and 11 reciprocally regulated mRNAs within 10 kb of dynamically expressed lncRNAs, reciprocal relationships of lncRNA and mRNA levels was validated for the Mccc1 and Relb genes using in vitro lncRNA knockdown in C2C12 cells. Network analysis suggested a central role for lncRNAs in modulating NFkappaB- and CREB1-regulated genes during embryonic heart growth and identified multiple mRNAs within these pathways that are also regulated, but independently of lncRNAs. Cardiac polyadenylated RNA (mRNA and lncRNA) profiles were generated from C57BL/6J mouse hearts were generated on Illumina HiSeq 2000 instruments. 7 independent E13.5 hearts, 12 adult hearts (6 at 6 weeks of age, 6 at 16 weeks of age), 4 sham-operated hearts at 12 weeks of age, and 4 hearts after 4 weeks of pressure overload (TAC) at 12 weeks of age.

Project description:Background: Frozen embryo transfer (FET) significantly upregulates fetal weight compared with fresh embryo transfer. However, the mechanism of FET increasing fetal weight remains unclear. Methods: We transferred blastocysts which had been vitrifified and warmed or fresh blastocysts into individual pseudopregnant recipients (n=11 mice each group) produced by natural mating to eliminate the influence of superovulation. The fetal weight, placental weight, placental efficiency and placenta architecture were compared. We performed placental lncRNA sequencing and mRNA sequencing to study the placental transcriptoms changes. And 15 selected differentially expressed lncRNAs and 15 mRNAs was checked. Findings: Enhanced birthweight (1.513 g vs. 1.685 g, P <0.001) was found in FET mice, with increased placental efficiency (11.355 vs. 10.312, P =0.045) and proportion of labyrinth zone area (59.78% vs. 53.19%, P = 0.034), indicating FET might change the placental function and morphology architecture. A total of placental 97608 mRNAs and 103339 lncRNAs were obtained, among which 1012 mRNAs and 554 lncRNAs showed significantly different expression between two groups. KEGG and GO enrichment analyses showed that the differentially expressed lncRNAs and their targeted mRNAs were related to labyrinth zone formation and maternal-fetal interface vascular remodeling. qRT-PCR, Western blotting and correlation analysis demonstrated that suppressed Lncenc1 to elevate Gjb5 might play a role in increased fetal weight of FET. Interpretation: FET changes the placental function and architecture resulting in higher mouse fetal weight, potentially via the network diagram of Lncenc1-miRNA-Gjb5.
