Novel Small RNA Modulates Virulence in Francisella tularensis

Project description:Most sRNAs control the expression of target genes by interacting with their mRNA. The fvr1 (Francisella virulence RNA 1) gene is found in the IGR between FTL_0777 and FTL_0778 and its sequence does not overlap those of the flanking genes, indicating that its target(s) is located in trans. Base-pairing between a sRNA and target mRNA generally leads to changed translation and often the stability of the mRNA is affected as well. Therefore, to identify possible targets of Fvr1, we compared the transcriptomes of the LVS?fvr1(LVS, for Live Vaccine Strain) and LVS/pfvr1+ strains after growth in liquid broth.

Project description:To understand differences of gene expression profiles between Francisella strains RNA profiles of Francisella strains were generated by deep sequencing, in triplicate, using NovaSeq6000. qRT–PCR validation was performed using SYBR Green assays. Our study represents the first detailed differential transcriptomic analysis of Francisella strains , with biologic replicates, generated by RNA-seq technology.

An AraC/XylS Family Transcriptional Regulator Modulates the Oxidative Stress Response of Francisella tularensis

Project description:Francisella tularensis is a Gram-negative bacterium that causes a fatal human disease known as tularemia. The Centers for Disease Control have classified F. tularensis as Category A Tier-1 Select Agent. The virulence mechanisms of Francisella are not entirely understood. Francisella possesses very few transcription regulators, and most of these regulate the expression of genes involved in intracellular survival and virulence. The F. tularensis genome sequence analysis reveals an AraC (FTL_0689) transcriptional regulator homologous to the AraC/XylS family of transcriptional regulators. In Gram-negative bacteria, AraC activates genes required for L-arabinose utilization and catabolism. The role of the FTL_0689 regulator in F. tularensis is not known. In this study, we characterized the role of FTL_0689 in gene regulation of F. tularensis and investigated its contribution to intracellular survival and virulence. The results demonstrate that FTL_0689 in Francisella is not required for L-arabinose utilization. Instead, FTL_0689 specifically regulates the expression of the oxidative and global stress response, virulence, metabolism, and other key pathways genes required by Francisella when exposed to oxidative stress. The FTL_0689 mutant is attenuated for intramacrophage growth, and mice infected with the FTL_0689 mutant survive better than wild-type F. tularensis LVS infected mice. Based on the deletion mutant phenotype, FTL_0689 was termed osrR (oxidative stress response regulator). Altogether, this study elucidates the role of the osrR transcriptional regulator in tularemia pathogenesis.

Characterization of inner and outer membranes proteins from Francisella tularensis strains LVS and Schu S4 and evaluation of possible vaccine candidates.

Project description:The inner and outer membranes were isolated from three biological replicates of two different strains of Francisella tularensis (LVS and Schu S4). Peptides were generated by proteolytic digestion with trypsin. Samples were analyzed on a quadrupole time-of-flight TripleTOF 5600 mass spectrometer and the data searched using Protein Pilot. Weighted spectral counting was performed using peptides that were at least 99% confident. Additionally, the membrane preparations were used to inoculate mice against a challenge with either LVS or SchuS4 strains. Immunoreactive bands from the mouse vaccination study were also extracted, digested with trypsin, and the spectral counts calculated.
