Concurrent progress of reprogramming and gene-correction to overcome limitation of gene therapy of ALK2 mutant iPSCs

ABSTRACT: To ameliorate the cumbersome processes of reprogramming and subsequent gene-editing in vulnerable iPSCs, we have developed a greatly simplified one-step procedure, simultaneously introducing reprogramming and gene-editing components into human fibroblast cells. This not only serves to save time, labor, and costs, but opens up a new arena of research that is beyond the current application of gene-editing methodologies due to restrictive reprogramming concerns, inhibitory pluripotency maintenance requirements, and vulnerability of single-cell dissociated iPSCs.

We generated iPSCs (mALK2-iPSCs) derived from Fibrodysplasia ossificans progressiva (FOP)-fibroblast cells carrying the ACVR1 p.R206H mutation and gene-corrected ALK2-iPSCs (cALK2-iPSCs). To identify gene-correction effects on global gene expression, gene expression profiling was measured in cALK2-iPSCs and mALK2-iPSCs, calibrated to WT-iPSCs with duplication.

Genetic stability in FA cells before and after gene editing and cell reprogramming.

Project description:Fibroblasts from a Fanconi anemia (FA) patient (FA-52) before and after correction by gene editing and transduction with a lentiviral vector expressing telomerase (geFA-52T fibroblasts). IPCs were generated from fibroblasts corrected by gene editing using STEMCCA LV (geFA-52T IPSCs clone 16) and finally the reprogramming cassette was excised (excised geFA-52T IPSCs clone 16.1) Four groups: Fibroblasts from a FA patient (FA-52) before and after correction by gene editing and transduction with a lentiviral vector expressing telomerase (geFA-52T) and gene editied IPSCs before and after excision of the reprogramming cassette (geFA-52T IPSCs clone 16 and excised geFA-52T IPSCs clone 16.1)

Phenotypic Correction of Oligodendrocytes by Concurrent Direct Reprogramming and Gene Replacement of Fibroblasts

Project description:Transcription factor-mediated direct reprogramming technologies that can convert one differentiated somatic cell to another, without an intermediate pluripotent stem cell stage, provide a new mechanism to access somatic cell types for disease modeling and possible therapeutic application. Previous efforts have focused on wild-type cells harboring transgenic reporters to signal successful reprogramming events. Outstanding questions remain of whether cells from disease backgrounds without transgenic reporters are capable of direct reprogramming and whether they phenocopy disease pathology. Here we demonstrate that fibroblasts from the shiverer mouse model of myelin disease can be directly reprogrammed to induced oligodendrocyte progenitor cells (iOPCs) by the expression of Sox10, Olig2, and Nkx6.2 without the aid of transgenic selection reporters. Shiverer mice harbor a homozygous ~20-kilobase deletion in the Myelin Basic Protein (Mbp) gene and this substantial deletion leads to abnormal oligodendrocytes resulting in hypomyelination and decreased lifespan. Shiverer iOPCs efficiently differentiated to induced oligodendrocytes (iOLs) which recapitulated the known disease phenotype of impaired axonal ensheathment and myelination capacity. Combining direct reprogramming and genetic supplementation of a minigene carrying regulatory elements and an open reading frame of MBP, restored MBP expression in differentiated iOLs and their ability to track along and ensheath microfibers in an in vitro assay of myelination capacity. Collectively these results show that iOPCs provide a rapid and accessible system to accurately model myelin disease pathology and provide promise for future therapeutic advancement for genetic myelin disorders.

Universal correction of blood coagulation factor VIII in patient-derived induced pluripotent stem cells using CRISPR/Cas9

Project description:Confirmation of the nuclease acivities at the On- and Off-target sites; Hemophilia A (HA) is caused by genetic mutations in the blood coagulation factor VIII (FVIII). Genome editing approaches can be used to target the mutated site itself in patient-derived induced pluripotent stem cells (iPSCs). However, these approaches can be hampered by difficulty preparing thousands of editing platforms for each corresponding variant found in HA patients. Here, we report a universal approach to correct the various mutations in HA patient iPSCs by the targeted insertion of the FVIII gene into the human H11 site via CRISPR/Cas9. We derived corrected clones from two types of patient iPSCs with frequencies of up to 64% and 66%, respectively, without detectable unwanted off-target mutations. Moreover, we demonstrated that endothelial cells differentiated from the corrected iPSCs successfully secreted functional protein in vitro and functionally rescued the disease phenotype in vivo. This strategy may provide a universal therapeutic method for correcting all genetic variants found in HA patients.

Correction of a Factor VIII genomic inversion with designer recombinases

Project description:Despite advances in nuclease-based genome editing technologies, correcting human disease-causing genomic inversions remains a challenge. Here, we describe the potential use of a recombinase-based system to correct a 140 kb int1h inversion frequently found in patients diagnosed with Hemophilia A. With the use of directed molecular evolution, we developed a linked heterodimeric recombinase system (RecF8) achieving 30% inversion of the target sequence in human tissue culture cells. Transient RecF8 treatment of endothelial cells, differentiated from int1h patient derived iPSCs, resulted in prominent correction of the inversion and restored Factor VIII mRNA expression. Our data suggest that the development of designer-recombinases represent an efficient and specific mean towards treatment of large gene inversions causing monogenic diseases.
