Project description:To further study the transcriptome of THP-1 human monocytes after exposure to of S-nitrosoglutathione (GSNO), we investigate whole genome microarray expression to identify genes regulated by exposure to GSNO (1.4 or 6 µM). Changes in gene expression in THP-1 cells incubated without (control) or with (1.4 or 6 µM) of GSNO for 4 h were measured. Four biological replicates were performed as controls: C1; C2; C3; C4. Four biological replicates were performed in 1,4 or 6 µM GSNO-exposed cells: G1; G2; G3; G4 and G5; G6; G7; G8; respectively.

Project description:Entamoeba histolytica is a protozoan parasite responsible for amebiasis, a disease which is characterized by acute inflammation of the colon. Adaptation of the parasite to toxic level of nitric oxide (NO) produced by phagocytes may be essential for the establishment of chronic amebiasis and for the survival of parasite within the host. To obtain insight into the mechanism of adaptation to NO, E. histolytica trophozoites were selected in vitro by stepwise exposures to increasing amounts of NO donor S-Nitrosoglutathione (GSNO) up to a concentration of 110 µM. These NO adapted trophozoites (NAT) were more resistance to acute exposure of GSNO (350μM), to activated macrophages and have a better capability to invade porcine colon explants compared to wild-type trophozoites. The transcriptome of NAT was investigated by RNA-sequencing (RNA-seq) and it showed only a weak overlapping with the transcriptome of trophozoites exposed to acute nitrosative stress (TEANS).

Project description:Pseudomonas extremaustralis, an Antarctic bacterium, was grown at low oxygen conditions for 24h and then exposed to S-Nitrosoglutathione (GSNO)100 µM for 1 h. RNA from treated and control samples was isolated using the Trizol method. RNA quality was analyzed on the Agilent Bioanalyzer and rRNA depletion was performed using the RiboZERO kit (Illumina). Samples were validated using an Agilent 2100 Bioanalyzer (Agilent Technologies). Libraries were prepared with TruSeq RNA Library Prep Kit v2 (Illumina) and sequenced with the Illumina NextSeq 550 platform with a single-end protocol.

Project description:Nitric oxide (NO) is a signaling molecule that participates in diverse developmental processes in plants, and in responses to abiotic and biotic stresses. The main mode of action of NO is protein S-nitrosylation, the post-translational modification of thiol residues. S-nitrosoglutathione (GSNO) as a biological NO donor has a very important function in NO signaling since it can transfer its NO moiety to other proteins. The breakdown of GSNO and thus the level of S-nitrosylated proteins are regulated by GSNO-reductase (GSNOR). Arabidopsis knock out gsnor mutants have a pleotropic phenotype with a developmentally differences and impaired responses to biotic and abiotic stresses. Therefore, the main aim of the experiment was to identify genes involving in NO signaling by comparing WT and gsnor mutant.
