Transcription profiling of mechanically stretched mouse NIH 3T3 cells to gain insights into the molecular and genetic basis of effect of mechanical stress

Project description:Type of experiment: Our experiments examine gene expression profiles from untreated and mechanical stretching NIH 3T3 cells to gain new insights about the molecular and genetic basis of effect of mechanical stress. Experimental factors: We compared differences in gene expression profiles between 0.25% mechanical strain and control factor for NIH 3T3 cells. The number of hybridizations performed in the experiment: A total of 4 hybridizations were performed on Agilent One-color microarray platform. Number of replicates (Biological or Technical) 3 biological replicates for treated condition Hybridization design: All treated NIH 3T3 cells sample RNAs were hybridized against a pooling control RNA sample (see above). Cells were cultured in a humidifier incubator at 37 0C and 5% CO2. After cells were confluent, these cells were subjected to cyclic sinusoidal strain at 0.25 Hz for 60 minutes at a maximum membrane indentation of 1 mm., equal to 0.25% strain. Strain experiments were carried out in a humidified incubator with 5% CO2 at 37°C. Control cells were treated identical, however, not exposed to mechanical stress.

Project description:In order to investigate the effects of downregulated autophagy in TM cells response to mechanical stretch, we conducted gene expression analysis in human TM cells deficient in autophagy and subjected to cyclic mechanical stretch. For this, three independent strains of primary human TM cells were transfected with a cocktail of siRNAs to specifically silence the expression of the autophagy genes Atg5 and Atg7 (siAtg5/7), and subjected to cyclic mechanical stress (15% elongation, 1 cycle/sec, 24h).

Comprehensive and comparative analysis of gene expression induced by LPA- and CP-609,550 in normal human epidermal keratinocytes (NHEKs)

Project description:We screened highly expressed G-protein coupled receptors (GPCRs) in normal human epidermal keratinocytes (NHEKs). We then tested the effect of ligands of selected GPCRs for their potency to induce the expression of FLG, a crucial skin barrier marker gene, in NHEKs. We used CP-609,550, a JAK inhibitor, as a positive control. As a result, we identified Oleoyl-L-α-lysophosphatidic acid (LPA) as a strong FLG inducer in NHEKs. In addition, we found that LPA induced NHEKs morphological changes characteristics of differentiated keratinocytes. Notably, although CP-609,550 also induced FLG expression in NHEKs, it didn't affect NHEKs cell morphology. We further conducted comprehensive and comparative microarray analysis of gene expression induced by LPA and CP-609,550 in NHEKs. We found that LPA treatment not only induced FLG expression but also expression of several genes related to keratinocytes differentiation, epidermis development and cornified envelope. This was not observed in the CP-609,550-treated cells.

Effect of Cyclic stretching on the gene expression in human spinal ligament cells

Project description:Ossification of the posterior longitudinal ligament (OPLL) of the spine is characterized by progressive ectopic bone formation in the spinal ligament. To identify the genes related to ectopic ossification of human spinal ligament affected by mechanical stress, analyses using cDNA microarray were carried out using cultured human spinal ligament cells that had been subjected to uniaxial cyclic stretching. cDNA microarrays revealed that ranges of distribution of both up- and down-regulated genes evoked by cyclic stretching were significantly wider in cells from than in non-OPLL cells. Project description:Analysis of undifferentiated keratinocytes or differentiated keratinocytes stimulated with or without human cathelicidin antimicrobial peptide (CAMP) LL37. Results provide insight into the biological effects of CAMP on human keratinocytes. NHEKs were divided into two groups; low calcium (0.05 mM) and high calcium condition (1.6 mM). Then keratinocytes were stimulated with human cathelicidin antimicrobial peptide LL37 at 0, 2.56, and 7.68 M-NM-<M for 12 h to 24 h.

Expression profiling of mechanical stretched interfollicular epidermal stem cells (IFESCs)

Project description:The renewing human epidermis constantly senses and adapts to a wide range of mechanical cues that are ubiquitous throughout life. The mechanisms of how mechanical forces are responded by interfollicular epidermal stem cells (IFESCs) and are transmitted directly into nucleus to modify gene expression remain incompletely defined. In vitro, human IFESCs were cultured on the collagen I coated silicon rubber membrane and then subjected to the mechanical stretched. Cyclic mechanical tension at 0.5 Hz sinusoidal curve at 10% elongation was applied using an FX-5000T™ Flexercell® Tension Plus™ unit (Flexcell International Corporation). In mechanical unloading groups, cells were cultured on the same plates in the same incubator with the mechanical stretched groups but not subjected to stretch. Combining genome-wide microarray and functional analyses, we made transcriptome analysis of samples from the mechanical unstretched or stretched isolated human IFESCs.

Indian hedgehog (Ihh) and its related signaling are key factors in normal enchondral ossification. However, the relation of OPLL to Ihh is unclear. The purpose of this study is to investigate the contribution of mechanical strain to OPLL and the relation of Ihh to OPLL. Cultured posterior longitudinal ligament cells were subjected to 24 hours of cyclic tensile strain and then analyzed by microarray.

Gene expression changes under cyclic mechanical stretch in rat retinal Muller cells

Project description:The retina is often subjected to tractional forces in a variety of conditions, for instance, pathological myopia, proliferative vitreoretinopathy. As the predominant glial element in the sensory retina, Muller cells are responsible for the homeostatic and metabolic support of retinal neurons and active players in virtually all forms of retinal injury and disease. Besides, Muller cells span the entire retinal thickness, extending from the inner to the outer limiting membranes, with cell bodies located in the inner nuclear layer and lateral processes expanding into the plexiform layers of the tissue. Because of this unique morphology, Muller cells can sense even minute changes in the retinal structure because of the mechanical stretching of their long processes or side branches. Thus, itM-bM-^@M-^Ys reasonable to infer that Muller cells also participate in ocular diseases when the retina is overstretched. In this study, we aim to investigate the whole genome regulation of Muller cells under mechanical stretching, which may help in excluding possible molecular mechanisms that would account for many retinal diseases in which the retina is often subjected to mechanical forces. We used microarrays to identify patterns of gene expression changes induced by cyclic mechanical stretching in Muller cells. Rat Muller cells were seeded onto flexible bottom culture plates and subjected to a cyclic stretching regimen of 15% equibiaxial stretching for 1 and 24 h.Muller cells cultured under the same conditions but with no applied mechanical strain were considered as the unstretched control. At each time points (1 and 24 h), three totally independent experiments (3 stretched samples and 3 control samples) were conducted.
