Project description:We have employed whole genome microarray to identify changes in gene expression in human bone marrow MSCs (hMSC-TERT) sorted from seven days co-cultures with breast cancer cell lines (MCF7) and (BT-20) Human MSC line (hMSC-TERT) were co-cultured with breast cancer cell lines MCF7 and BT-20 for seven days then were sorted using EpiCAM MACS beads. Subsequently, RNA was extracted using Norgen total RNA isolation kit. Extracted RNA was labeled and then hybridized to the one-color Agilent Human GE 8x60K Microarray chip.

Project description:We aimed in this study to identify the differentially regulated genes by Dlk1 in hMSC cells using microarray technology in order to gain a better understanding of Dlk1-mediated signaling pathways during hMSC differentiation. Both control (hMSC-TERT)(not expressing Dlk1) and Dlk1 overexpressing cells (hMSC-Dlk1) were cultured in triplicate at 3×104 cells/cm2 in Petri-dish in standard growth medium. At 90-100% confluence, highly purified total cellular RNA was isolated from each of three independent cultures per cell line using RNeasy Kit (QIAGEN Nordic, West Sussex, UK) according to the manufacturer?s instructions

The samples were labeled using the miRCURY LNAM-bM-^DM-" microRNA Hi-Power Labeling Kit, Hy3M-bM-^DM-"/Hy5M-bM-^DM-" and hybridized on the miRCURY LNAM-bM-^DM-" microRNA Array (6'th GEN). Single-cell full-length RNA-sequencing of MCF7 and BT-474 cells using down-scaled Smart-seq2

Project description:We report single-cell RNA-sequencing data of wild type, breast cancer cells MCF7 and BT-474. Single cells were isolated using the F.SIGHT single cell dispenser. cNDA synthesis and library preparation were down-scaled to 1/10 of original reaction volumes. By comparative UMAP clustering we could show that our MCF7-cluster exactly overlapped with sequencing data from GSE151334. Data was analyzed using three common alignment tools: salmon, kallisto and STAR. DE analysis was performed with inputs from each aligner. We proved the overexpression of ErbB2 in BT-474 compared to MCF7 in all cases.
