A transcriptome map of Actinobacillus pleuropneumoniae at single-nucleotide resolution using RNA-seq

ABSTRACT: We used the RNA-seq technology to do a genome-wide transcriptional analysis of A. pleuropneumoniae strain JL03 and investigated the transcriptome structure at a single-nucleotide resolution.The RNA-Seq based transcriptome map validated annotated genes and corrected annotations of open reading frames in the genome, and led to the identification of many functional elements (e.g. regions encoding novel proteins, non-coding sRNAs and operon structures).

Transcription profiling of Actinobacillus pleuropneumoniae wild type, hns mutants and rseA mutants

Project description:Clinical isolates of the porcine pathogen Actinobacillus pleuropneumoniae often form adherent colonies on agar plates due to expression of an operon, pgaABCD, encoding a poly-N-acetylglucosamine (PGA) extracellular matrix. The adherent colony phenotype, which correlates with the ability to form a biofilm on the surface of polystyrene plates, is lost following serial passage in broth culture, and repeated passage of the non-adherent variants on solid media does not result in reversion to the adherent colony phenotype. In order to investigate the regulation of PGA expression and biofilm formation in A. pleuropneumoniae, we screened a bank of transposon mutants of the non-adherent serovar 1 strain, S4074T, and identified mutations in two genes, rseA and hns, which resulted in formation of the adherent colony phenotype. In other bacteria, including the Enterobacteriaceae, H-NS acts as a global gene regulator, and RseA is a negative regulator of the extracytoplasmic stress response sigma factor, ?E. Transcription profiling of A. pleuropneumoniae rseA and hns mutants revealed that both ?E and H-NS independently regulate expression of the pga operon. Transcription of the pga operon is initiated from a ?E promoter site in the absence of H-NS, and up-regulation of ?E is sufficient to displace H-NS, allowing transcription to proceed. In A. pleuropneumoniae, H-NS does not act as a global gene regulator, but rather specifically regulates biofilm formation via repression of the pga operon. Positive regulation of the pga operon by ?E indicates that biofilm formation in is part of the extracytoplasmic stress response in A. pleuropneumoniae.

Expression data of Actinobacillus pleuropneumoniae 4074 and the ΔluxS mutant of Actinobacillus pleuropneumoniae 4074

Project description:LuxS is an enzyme involved in the activated methyl cycle and the by-product autoinducer 2 (AI-2) was a quorum sensing signal in some species. In our previous study, the functional LuxS in AI-2 production was verified in the porcine respiratory pathogen Actinobacillus pleuropneumoniae. Enhanced biofilm formation and reduced virulence were observed in the luxS mutant. To comprehensively understand the luxS function, in this study, the transcriptional profiles were compared between the A. pleuropneumoniae luxS mutant and its parental strain in four different growth phases using microarray. Many genes associated with infection were differentially expressed. The biofilm formation genes pgaABC in the luxS mutant were up-regulated in early exponential phase, while 8 genes associated with adhesion were down-regulated in late exponential phase. A group of genes involved in iron acquisition and metabolism were regulated in four growth phases. Further investigations on these virulence traits demonstrated that the luxS mutant showed enhanced biofilm formation and reduced adhesion ability and these effects were not due to lack of AI-2. But AI-2 could increase biofilm formation and adhesion of A. pleuropneumoniae independent of LuxS. Growth under iron restricted condition could be controlled by LuxS through AI-2 production. These results revealed pleiotropic roles of LuxS and AI-2 on A. pleuropneumoniae virulence traits.

Project description:Porcine pleuropneumonia caused by Actinobacillus pleuropneumoniae affects pig health status and the swine industry worldwide. Despite of the extensive number of studies focused on A. pleuropneumoniae infection and vaccine development, its exoproteome is still rather unexplored. By combining high-throughput mass spectrometry and immunoproteomic approaches, with our current work we provide an in-depth characterisation of A. pleuropneumoniae serotype 2 exoproteome. Label-free liquid chromatography - tandem mass spectrometry (LC-MS/MS) combined with a comprehensive bioinformatics analysis revealed 484 secreted proteins, of which 84 were predicted to be virulence factors and 142 to be exported via different export mechanisms. The RTX toxins ApxIIA, ApxIIIA and ApxIVA were found to be the most abundant proteins in the A. pleuropneumoniae serotype 2 exoproteome, although ApxIVA is commonly assumed to be expressed exclusively in vivo. Immunoproteomic approaches coupled to LC-MS/MS analysis allowed to portray the immunogenic proteins within the bacterial exoproteome to identify potential vaccine candidates. Using serum pools from uninfected, acutely infected and chronically infected animals, we were able to monitor the seroconversion during disease progression. Overall, our work is expected to contribute to the understanding of the complex pathogenic mechanisms and to facilitate the discovery of potential antimicrobial agents for controlling porcine pleuropneumonia.
