Project description:Clinical evidence supports the occurrence of intermittent diarrhea in many type 1 diabetes mellitus (T1DM) patients. Others and we found that net fluid absorption rate is dramatically lower in the ileum of T1DM murine models. However, the identity of molecules that contribute to fluid malabsorption in the gut remains unknown. Considering the importance of ion transporters and channels for intestinal fluid absorption, we reasoned that their expression level at the luminal membrane is altered under diabetic conditions. To this end, we analyzed the brush border membrane vesicles (BBMVs) from the ileum of control and DM mice by proteomic analysis. The expression levels of Cl-/HCO3- exchanger SLC26A3/DRA and cystic fibrosis transmembrane conductance regulator (CFTR) were not significantly different between control and DM mice. Cl-/HCO3- exchanger Slc26a6/PAT1 and Na+/H+ exchanger 3 (NHE3) were not detected. Interestingly, cytoskeleton scaffold proteins that regulate NHE3, including NHERF1-3 and ezrin, were all significantly lower in diabetic animals.

Project description:The sodium chloride cotransporters (NCC) and the epithelial sodium channel (ENaC) have been reported to be the main effectors of aldosterone in Na+ and Cl- reabsorption in the distal nephron and dietary K+ downregulates NCC. The Cl-/HCO3- exchanger pendrin has been revealed as a key role in maintaining extracellular fluid and electrolyte homeostasis by regulating Cl- reabsorption in the aldosterone-sensitive distal nephron working in tandem with ENaC and perhaps NCC. Pendrin-mediated Cl-/HCO3- exchange is stimulated in models of metabolic alkalosis (e.g. aldosterone administration or dietary NaHCO3 intake), and plasma K+ level is thought to negatively influence pendrin. In this study we used high-throughput proteomics and bioinformatics approaches to analyze urinary exosome contents isolated from patients undergoing fludrocortisone suppression test (FST, as a means of confirming or excluding primary aldosteronism [PA]) to examine the actions of administration of aldosterone analogs, NaCl and KCl orally on transmembrane proteins existing in urinary exosomes.

Project description:Na+/H+ exchanger 1 (NHE1; SLC9A1) is a transport protein responsible for pH regulation in various types of cells. In cardiac myocytes, NHE1 generates the largest transmembrane acid-extrusion flux and is therefore the most significant controller of the intracellular acid/base milieu. The protein itself is regulated by various enzymes, including kinases. A number of critically important sites have been identified, coupling NHE1 to signalling cascades such as cAMP. NHE1 activity is very sensitive to oxygen tension, thus linking metabolism with pH. However, the sites implicated in this effect are not known. This study identified novel phosphorylation sites on NHE1. The state of these residues regulates NHE1 activity, hence pH and a myriad of pH sensitive downstream processes. Dysregulated NHE1 activity has been shown in diseases of the heart. Identification of novel regulatory sites can help demarcate the mechanisms of disease and suggest possible interventions to correct NHE1 activity.

Project description:Skeletal muscle is a major site of postprandial glucose disposal. Inadequate insulin action in this tissue contributes to hyperglycemia in type 1 and type 2 diabetes. While glucose is known to stimulate insulin secretion by pancreatic b cells, whether it directly engages nutrient-sensing pathways in skeletal muscle to regulate glucose metabolism remains largely unexplored. Here we identified the Baf60c-Deptor-AKT pathway as a target of muscle glucose sensing that augments insulin action in skeletal myocytes. Genetic activation of this pathway improves postprandial glucose disposal in mice, whereas the muscle-specific ablation impaired insulin action and led to glucose intolerance. Mechanistically, glucose triggers a rapid calcium response in myocytes that acts on the class IIa histone deacetylase HDAC5, leading to Baf60c induction and insulin-independent AKT activation. The pathway is engaged by the anti-diabetic drugs sulfonylureas, suggesting that its therapeutic activation may have beneficial effects on glycemic control in diabetes. We used microarrays to elucidate the role of of Baf60c in transcriptional regulation by glucose in skeletal myocytes.

Project description:Patients with short QT syndrome (SQTS) may present with syncope, ventricular fibrillation or sudden cardiac death. Six SQTS susceptibility genes, encoding cation channels, explain less than 25% of SQTS cases. Here we identify a missense mutation in the anion exchanger (AE3)-encoding SLC4A3 gene in two unrelated families with SQTS. The mutation causes reduced surface expression of AE3 and reduced membrane bicarbonate transport. Slc4a3 knockdown in zebrafish causes increased cardiac pHi, short QTc, and reduced systolic duration, which is rescued by wildtype but not mutated SLC4A3. Mechanistic analyses suggest that an increase in pHi and decrease in [Cl-]i shortened the action potential duration. However, other mechanisms may also play a role. Altered anion transport represents a mechanism for development of arrhythmia and may provide new therapeutic possibilities.

Project description:Mutations in ClC-5 cause Dent's disease, a disorder associated with low molecular weight proteinuria, hyperphosphaturia and kidney stones. ClC-5 is a Cl /H+-exchanger predominantly expressed in the kidney, where it facilitates the acidification of proximal tubular endosomes. Project description:The purpose of this study was to investigate the hypothesis that cardiomyocyte-specific loss of the electrogenic NBCe1 Na+-HCO3- cotransporter is cardioprotective during in vivo ischemia-reperfusion (IR) injury. An NBCe1 (Slc4a4 gene) cardiac conditional knockout mouse (KO) model was prepared by gene targeting. Cardiovascular performance of wild-type (WT) and cardiac-specific NBCe1 KO mice was analyzed by intraventricular pressure measurements, and changes in cardiac gene expression were determined by RNA Seq analysis. Response to in vivo I/R injury was analyzed after 30 minutes occlusion of the left anterior descending artery followed by 3 hours of reperfusion. Loss of NBCe1 in cardiac myocytes did not impair cardiac contractility or relaxation and caused only limited changes in gene expression patterns, such as those for electrical excitability. However, following ischemia and reperfusion, KO heart sections exhibited significantly fewer apoptotic nuclei than WT sections. These studies indicate that cardiac-specific loss of NBCe1 does not impair cardiovascular performance, causes only minimal changes in gene expression patterns, and protects against I/R injury in vivo.

Project description:IRBIT is a multifunctional protein that controls the activity of IP3 receptors and the transporters NBCe1-B, CFTR and Slc26a6, by an unknown mechanism. IRBIT interacts with the NBCe1-B N terminus autoinhibitory domain (AID), exposing two cryptic Cl- sensing GXXXP sites to confer regulation of NBCe1-B by intracellular Cl- (Cl-in). Here, LC-MS/MS phosphoprotein analysis revealed that IRBIT controls five NBCe1-B phosphorylation sites that determine both NBCe1-B active conformation and regulation by Cl-in. Specifically, IRBIT-dependent combinatorial dephosphorylation of pSer232, pSer233 or pSer235 generated the conformations pSer233/pSer235, pSer232/pSer235 or pSer232/pSer233. The activity of the pSer232/pSer235 form is similar to IRBIT-activated NBCe1-B, but it is insensitive to inhibition by Cl-in. The pSer232/pSer235 form properties were similar to wild-type NBCe1-B, while the pSer232/pSer233 form was partially active, further activated by IRBIT, but retained inhibition by Cl-in. In addition, IRBIT recruited the phosphatase PP1 and the kinase SPAK to control Ser65 phosphorylation, which reciprocally affected Cl-in sensing by the 32GXXXP36 motif. IRBIT also recruited the phosphatase calcineurin and the kinase CaMKII to control phosphorylation of Ser12, which reciprocally affected Cl-in sensing by the 194GXXXP198 motif. Ser232/Ser233/Ser235 are conserved in all NBCe1 variants. Mutating the serines to alanines resulted in fully active NBCe1 transporters, while mutating the serines to the phosphomimetic aspartates resulted in inhibited transporters, suggesting that Ser232/Ser233/Ser235 determine whether NBC transporters are in active or inactive conformations. These findings reveal how multiple kinase/phosphatase pathways use multiple phosphorylation sites to fine tune a transport function, which have important implications for epithelial fluid and HCO3- secretion.

Project description:Zebrafish ncx1h (also known as slc8a1a) encodes a cardiac specific sodium-calcium exchanger 1 (NCX1h), a primary Ca 2+ efflux effector in cardiomyocytes. The zebrafish tremblor (tre) mutant lacks functional NCX1h and, consequentially, cyclic Ca2+ transients are abolished. In this study, we investigated the effect of aberrant Ca2+ homeostasis in cardiomyocytes on gene expression. Wild type and ncx1 mutant hearts at 48 hours post fertilization were isolated for RNA preparation and gene expression analysis was performed using an Affymetrix Zebrafish GeneChip.
