Project description:Proteins and peptides are minor components of vegetal oils. The presence of these compounds in virgin olive oil was first reported in 2001, but the nature of the olive oil proteome is still a puzzling question for food science researchers. In this project, we have compiled for a first time a comprehensive proteomic dataset of olive fruit and fungal proteins that are present at low but measurable concentrations in a vegetable oil from a crop of great agronomical relevance as olive (Olea europaea L.). Accurate mass nLC-MS data were collected in high definition direct data analysis (HD-DDA) mode using the ion mobility separation step. Protein identification was performed using the Mascot Server v2.2.07 software (Matrix Science) against an ad hoc database made of olive protein entries. Starting from this proteomic record, the impact of these proteins on olive oil stability and quality could be tested. Moreover, the effect of olive oil proteins on human health and their potential use as functional food components could be also evaluated. In addition, this dataset provides a resource for use in further functional comparisons across other vegetable oils, and also expands the proteomic resources to non-model species, thus also allowing further comparative inter-species studies.

Project description:Olive oil is protective against risk factors for cardiovascular and cancer diseases. A nutrigenomic approach was performed to assess whether olive oil, the main fat of the Mediterranean diet modifies the gene expression in human peripheral blood mononuclear cells. Six healthy male volunteers ingested, at fasting state, 50 ml of olive oil, and continued with the same olive oil as a source of raw fat (25ml/day) during 3 weeks. Prior to intervention a 1-week washout period with sunflower oil as the only source of fat was followed. During the 3 days before, and on the intervention day, a very low phenolic compound diet was followed. At baseline (0h), at post ingestion (6h), and at fasting state after 3 weeks of sustained consumption of olive oil total RNA was isolated from PBMC. Project description:In this comprehensive study, we delve into the examination of the phenolic fraction found in Extra Virgin Olive Oils (EVOOs) and its potential impact on endothelial cells, with a particular focus on its role in angiogenesis. To accomplish this, we employed a phenolic extract derived from EVOO sourced from the Picual variety in Jaén, Spain. Our investigation of an extract offers a compelling advantage, enabling us to obtain larger quantities of bioactive molecules compared to conventional dietary consumption, provided that the concentrations of these diverse compounds within the mixture are deemed safe. Furthermore, this approach permits us to meticulously isolate and assess the effects of these molecules independently from the other components of EVOO, such as fatty acids and more.

Project description:SCOPE: A previous study demonstrated that intake of olive pomace extract containing maslinic acid (MA), a triterpene, effectively prevents and alleviates arthritis in animals and humans. Here, the molecular mechanisms involved in the anti-arthritis effect of MA have been elucidated by determining gene expression changes induced by olive-derived MA intake in collagen antibody-induced arthritis (CAIA) mice. METHODS AND RESULTS: Mice are divided into the untreated (CT), CAIA (CA), and CAIA administered MA (CA + MA) groups. The CA + MA mice are fed MA at a daily dose of 200 mg kg-1 of body weight from day 1. CAIA is then induced on day 8 and evaluated on day 12. Arthritis symptoms are alleviated, and the gene expression of inflammatory cytokines is reduced in the CA + MA group compared with the CA group. A DNA microarray analysis of synovial membranes reveals that MA alters the expression levels of genes related to inflammation, including glucocorticoid responses, immune responses, and the extracellular matrix. CONCLUSIONS: The preventive effect of MA on arthritis is attributable to the promotion of tissue formation as well as suppression of inflammation in the synovium via inactivation of Toll-like receptor signaling and downregulation of leukotrienes through the glucocorticoid receptor.

Project description:The aim of this study was to evaluate whether the dietary treatment with olive oil phenols modifies the profile of microRNA and gene expression in different areas of the aging mouse brain and to correlate these changes with the cognitive and motor changes observed in the same aging animals. C57Bl/6J mice were fed from middle age to senescence with extra-virgin olive oil (10% wt/wt dry diet) rich in polyphenols (H-EVOO group, total polyphenol dose/day, 6mg/kg) or with the same extra-virgin olive oil deprived of phenolic compounds (L-EVOO group). By whole gene expression analysis we identified 53 genes differentially expressed in the cortex of H-EVOO mice compared to L-EVOO mice and no genes differentially expressed in the cerebellum. Gene Set Expression analysis (GSEA) found 6 gene sets significantly modulated by the dietary treatments in cortex, like the agrin-related postsynaptic differentiation gene set, involved in cholinergic synaptic differentiation and maintenance, exerting effects on axonal and dendritic growth. miRNA profiling found only 6 miRNA differentially modulated after 12 months of feeding and 63 after six months. Among them we noted miRNAs previously associated to neurodegenerative disorders such as mir101, 8.5 time more expressed in the cortex of L-EVOO fed mice compared to the H-EVOO group and no expressed in young mice. We extended our analysis on cortex harvested from transgenic TgCRND8 mice, a model of Alzheimer's disease, observing an overall down-regulation of miRNAs compared to mice of the same age. On the contrary, the H-EVOO fed mice cortex showed expression profiles similar to those of young mice, results supporting our previous data demonstrating that extra virgin olive oil rich in polyphenols may improve some age-related dysfunctions.

Project description:Some studies have shown that acute intake of high-phenol virgin olive oil reduces pro-inflammatory, pro-oxidant and pro-thrombotic states, but nowadays still remains unclear if those effects attributed to its phenolic fraction could be exerted at transcriptional level in vivo. Two virgin olive oil-based breakfasts with high (398 ppm) and low (70 ppm) content of phenolic compounds were administered to twenty patients (9 men and 11 women) with metabolic syndrome following a double-blinded random crossover design. Prior to the first breakfast intervention, participants followed a 6-week washout period in which they were instructed to not take vitamins, soy supplements, or any drug. To eliminate the potential effect that might exist in their usual dietary habits, all subjects followed a low-fat, carbohydrate rich (CHO) diet during this period and untill the end of the study. In the 24 h before each breakfast intervention, participants were instructed to avoid consuming phenol-rich foods such as fruit or juices, wine, grape juice, chocolate, coffee, tea, olive oil, or soya, and to refrain from intense physical exercise during that period. After a 12 h fasting and following a randomized sequential crossover design with one-week washout period, participants reported to the hospital and received two fat meals consisting of 60 g of white bread, 40 mL of VOO (CANOLIVA®, Antonio Cano e Hijos™, Córdoba, Spain) with high (398 ppm) or low (70 ppm) content in phenolic compounds, and 60,000 IU of vitamin A per m2 of body surface. Olive oil with low content in phenolic compounds was obtained as a result of extraction by physical procedures of most of the phenolic compounds in the high-phenol olive oil, so that both oils kept a similar composition of the remaining macro- and micronutrients. Throughout the 4-h duration of the study, the subjects performed no physical activity, nor did they consume anything but water. Gene expression was performed on peripheral blood mononuclear cells (PBMCs) at postprandial period (4 hours after breakfast) by microarray analysis and verified by qRT-PCR.
