Project description:Arabidopsis ABA HYPERSENSITIVE1 (ABH1) / CAP-BINDING-PROTEIN80 functions in cap-binding, splicing, ABA and stress response signaling and in the biogenesis of microRNAs (miRNAs). SUPPRESSOR OF GENE SILENCING3 (SGS3) plays a crucial role in post transcriptional gene silencing (PTGS). We show that abh1 sgs3 double mutant manifests pleiotropic phenotypes including altered ABA sensitivity in seed germination, fused cotyledons, delayed growth in root initiation and leaf production, accelerated juvenile-to-adult phase transition and flowering time, short and bent siliques with ectopic carpelloid tissues, reduced seed set due to altered carpel structure and pollen tube growth, and increased resistance to powdery mildew infection. A systems approach using meta-analysis of whole genome tiling microarray datasets for mutants of PTGS, RNA processing, and responses to environmental stress was taken to design a 135k feature custom microarray to probe the molecular bases of these phenotypes. Results confirmed documented effects of ABA or abh1 on precursor MIRNA expression (pre-MIR159, pre-MIR168) and identified novel miRNAs and their targets regulated by ABA (pre-MIR408, LACCASE12 and -13; pre-MIR824) or elevated in abh1 sgs3 double mutant (pre-MIR156A,C; pre-MIR164B, pre-MIR167A, pre-MIR408, At5g15845/CONSTANS-LIKE1-cis-NAT-RNA). miR156 targets SQUAMOSA-PROMOTER-BINDING-PROTEIN-LIKE genes that spawn siRNAs and control phase change; miR164 targets CUP-SHAPED-COTYLEDON1 and -2 genes; miR166 methylates target PHABULOSA genomic DNA in trans; miR167 targets AUXIN RESPONSE FACTOR 6 (ARF6) and ARF8 which regulate both female and male reproduction; miR408 targets a plantacyanin which is expressed predominantly in the transmitting tract of the pistil and affects pollen tube guidance on the stigma. We also identified sgs3 by ABA interactions on gene expression which reveal potential nodes in regulatory networks linking ABA, mRNA processing and PTGS with development.

Project description:Abscisic acid (ABA) is a plant hormone that is important in responding to various environmental stresses. Using an ABA auxotroph in Arabidopsis (aba2-2) as the plant material, we would like to identify early downstream targets of transcription in response to a small dose of ABA, 1 uM. We also added cycloheximide to preferentially obtain immediate targets of ABA addition. We believe that using a sensitized background of an ABA auxotroph would yield a set of genes that are very closely regulated by ABA at the transcriptional level. This data set will be used for network analysis of ABA signaling.

Project description:The plant hormone abscisic acid (ABA) is best known for its role as a regulator of the responses to abiotic stressors. Components of ABA signaling pathway are thus considered as promising targets for securing yield under stress. ABA levels rise in response to abiotic stress, mounting a number of physiological and metabolic responses that promote plant survival under unfavorable conditions. ABA elicits its effects by binding to a family of soluble receptors called PYR/PYL/RCAR. Arabidopsis genome encodes 14 PYL proteins, knowledge on differential biological functions of these members is scarce. In this work, we took a gain-of-function approach to predict receptor-specific functionality. We introduced a set of activating mutations in the mobile gate and α-helix of sub family II ABA receptors. These mutations constitutively enforce the active ABA-bound conformation. We then transformed ABA deficient mutants with the constitutive receptors and monitored suppression of ABA deficiency phenotype. Our findings suggest that subfamily II receptors have differential activity in regulating transpiration and transcription of oxidation and stress response genes. These differences partly derived from differential accumulation of PYLs in the guard cells. Data mining revealed that only a small subset of PYL receptors are transcribed in the guard cells in a steady state. Because the guard cell specific receptors have different affinity to ABA and to the PP2C co-receptor we propose that, a combined partial receptor redundancy facilitate a gradual activation of the ABA output signal as its cellular concentration rises via biochemically different receptors.

Here we show that ABA induced phosphorylation of three basic helix-loop-helix (bHLH) transcription factors, called AKSs (ABA-RESPONSIVE KINASE SUBSTRATES; AKS1, AKS2, AKS3), in Arabidopsis guard cells, and that they facilitated stomatal opening through the transcription of genes encoding inwardly-rectifying K+ channels. aks1aks2-1 double mutant plants showed decreases in light-induced stomatal opening, K+ accumulation in response to light, activity of inwardly-rectifying K+ channels, and transcription of genes encoding major inwardly-rectifying K+ channels without affecting ABA-mediated stomatal closure. Overexpression of POTASSIUM CHANNEL IN ARABIDOPSIS THALIANA 1 (KAT1), which encodes a major inwardly-rectifying K+ channel in guard cells, rescued the phenotype of aks1aks2-1 plants. AKS1 bound directly to the promoter of KAT1, an interaction that was attenuated after ABA-induced phosphorylation. The ABA agonist pyrabactin induced phosphorylation of AKSs. Our results demonstrate that the AKS family of bHLH transcription factors facilitates stomatal opening through transcription of genes encoding inwardly-rectifying K+ channels, and that ABA suppresses the activity of inwardly-rectifying K+ channel activity by triggering the phosphorylation of these transcription factors.
