Transcriptome of mouse thymic and thyroid epithelial cells

ABSTRACT: We have compared the transcriptional profiles from two epithelial cell types to find genes specifically expressed in thymic medullary epithelial cells. Epithelial cells from thyroid and thymic medulla from C57BL/6 mice were sorted and their transcriptional profiles compared. Cells were gated as viable, Epcam+ CD45- for thyroid and viable, Epcam+ CD45- UEA+ for thymic medulla.

Project description:To investigate gene expression profile of medullary thymic epithelial cells with high surface density of MHC class II (mTEChigh), a murine parent into F1 hematopoietic stem cell transplantation model was administered. As recipient mice (B6xDBA/2)F1 (B6D2F1) were used that received either a syngeneic transplant from a (B6xDBA/2)F1 mouse or an allogeneic transplant from a B6 mouse, which leads to acute graft-versus-host disease (aGVHD). mTEChigh (CD45-EpCAM+Ly51-UEA1+MHCIIhigh) are sorted by FACS.

Meis1 deficiency effect on thymic epithelial cells

Project description:Analysis of thymic epithelial cells lacking Meis1 gene using K14-CreERT2 Meis1fl/fl mice. Meis1 is a TALE class homeodomain transcription factor that critically regulates numerous embryonic developmental processes. Results provide insight into the role of Meis1 in the maintenance of postnatal thymic epithelial cells. Meis1-regulated gene expression in CD45- EpCAM+ mouse thymic epithelial cells was measured at 4 days after the induction of Meis1 deletion. Two independent experiments (pools of 6 mice; 2 pools per genotype) were performed.

CD90 marks a mesenchymal program in human thymic epithelial cells in vitro and in vivo

Project description:Thymic epithelium is critical for the structural integrity of the thymus and for T cell development. Within the fully formed thymus, large numbers of hematopoietic cells shape the thymic epithelium into a scaffold-like structure which bears little similarity to classical epithelial layers, such as those observed in the skin, intestine or pancreas. Here, we show that human thymic epithelial cells (TECs) possess an epithelial identity that also incorporates the expression of mesenchymal cell associated genes, whose expression levels vary between medullary and cortical TECs (m/cTECs). Using pluripotent stem cell (PSC) differentiation systems, we identified a unique population of cells that co-expressed the master TEC transcription factor FOXN1, as well as the epithelial associated marker EPCAM and the mesenchymal associated gene CD90. Using the same serum free culture conditions, we also observed co-expression of EPCAM and CD90 on cultured TECs derived from neonatal human thymus in vitro. Single cell RNA-sequencing revealed these cultured TECs possessed an immature mTEC phenotype and expressed epithelial and mesenchymal associated genes, such as EPCAM, CLDN4, CD90 and COL1A1. Importantly, flow cytometry and single cell RNA-sequencing analysis further confirmed the presence of an EPCAM+CD90+ population in the CD45- fraction of neonatal human thymic stromal cells in vivo. Using the human thymus cell atlas, we found that cTECs displayed more pronounced mesenchymal characteristics than mTECs during embryonic development. Collectively, these results suggest human TECs possess a hybrid gene expression program comprising both epithelial and mesenchymal elements, and provide a basis for the further exploration of thymus development from primary tissues and from the in vitro differentiation of PSCs.

The thymoproteasome hardwires the TCR repertoire of CD8+ T cells in the cortex independent of negative selection

Project description:The thymoproteasome expressed specifically in thymic cortical epithelium optimizes the generation of CD8+ T cells. However, how the thymoproteasome contributes to CD8+ T cell development is unclear. Here we show that the thymoproteasome shapes the TCR repertoire directly in cortical thymocytes before migration to the thymic medulla. We further show that the thymoproteasome optimizes CD8+ T cell production independent of the thymic medulla, independent of additional antigen-presenting cells including medullary thymic epithelial cells and dendritic cells, and independent of apoptosis-mediated negative selection. These results indicate that the thymoproteasome hardwires the TCR repertoire of CD8+ T cells with cortical positive selection independent of negative selection in the thymus.
