Project description:Sucrose synthase (SUS), a key enzyme of sugar metabolism, plays an important role in the regulation of carbon partitioning in plant, and affects important agronomic traits and abiotic responses to adversity. However, the function of ZmSUS1 in plant drought tolerance is still unknown. In this study, the expression patterns of ZmSUS1 in different tissues and under drought stress were analyzed in maize (Zea mays L.). It was found that ZmSUS1 was highly expressed during kernel development but also in leaves and roots of maize, and ZmSUS1 was induced by drought stress. Homozygous transgenic maize lines of ZmSUS1 overexpressing increased the content and activity of SUS under drought stress and exhibited higher relative water content, proline and abscisic acid content in leaves. Specifically, the net photosynthetic rate and the soluble sugar contents including sucrose、glucose、fructose、UDP-glucose and ADP-glucose in transgenic plants was significantly improved after drought stress. RNA-seq analysis showed that overexpressing of ZmSUS1 mainly affected the expression level of carbon metabolism-related genes, especially the expression level of sucrose metabolism-related genes including ZmSUS、ZmSPS and ZmINV were significantly up-regulated in transgenic maize. Overall, these results suggested that ZmSUS1 improved drought tolerance by regulating sucrose metabolism and increasing soluble sugar content, and endowing transgenic maize with higher relative water content and photosynthesis levels, which can be served as a new gene candidate for cultivating drought resistant maize varieties.

Project description:Drought represents a major constraint on maize production worldwide. Understanding the genetic basis for natural variation in drought tolerance of maize may facilitate efforts to improve this trait in cultivated germplasm. Here, using a genome-wide association study, we show that a miniature inverted-repeat transposable element (MITE) inserted in the promoter of a NAC gene (ZmNAC111) is significantly associated with natural variation in maize drought tolerance. For maize RNA-seq analysis, pooled tissues from three, eight-day-old maize seedlings were collected from transgenic and wild-type plants, prior to or after 2-hour dehydration, to conduct the RNA-seq analysis.

Project description:In this study a transcriptomic approach (RNA-sequencing) was utilized to elucidate molecular responses of maize (Zea mays L.) primary roots of the inbred line B73 to water deficit to gain a better understanding of the mechanisms underlying drought tolerance. Kernels of the maize inbred line B73 were germinated in paper rolls soaked with distilled water until seedlings had a primary root length of 2 to 4 cm. For mild and severe water deficit conditions, seedlings were transferred to PEG8000 solution with water potentials of -0.2 MPa and -0.8 MPa, respectively. Water deficit treatment was applied for 6 h and 24 h. Each treatment was performed in four biological replicates each consisting of 10 roots.

Project description:During their lifespan, plants continuously respond to a multitude of stressful factors that affects their development and reproductive fitness through a sophisticated variety of physiological, biochemical, transcriptional, and epigenetic mechanisms. Dynamic changes in chromatin structure and concomitant transcriptional variations play an important role in both stress response and adaptation. Epigenetic memory mechanisms seem to be involved because chromatin and expression patterns can be indeed stably maintained during cell division, once the triggering stimulus has been removed. In this study we performed transcriptome and genome-wide analysis of histone modifications of maize plants subjected to a mild and prolonged drought stress followed by a complete recovery phase. Results led to the identification of stress-responsive genes in which variations in transcript and/or histone modification levels persist after stress removal. Genes not showing an immediate response to stress, but perceiving and storing stress signal for a delayed response were also identified. This provides valuable information about genes and mechanisms utilized by plants to adapt to stressful environment and potentially involved in stress memory. We also report that drought stress alters flowering and inflorescences patterning, possibly by affecting expression and chromatin of flowering regulatory genes.
