The effect of whole-glucan particles on the gene expression of polarized M2 bone-derived macrophages

ABSTRACT: Bone-marrow macrophages polarized to M2 phenotype are immunosuppressive. Interestingly, treatment with whole-glucan particles converts M2 macrophages to M1 phenotype with an anti-tumor phenotype. In this study, the effect of WGP treatment for 6 hours on the gene expression of M2 macrophages was assessed.

Project description:Analysis of the effects of CNI-1493 treatment on M1 and M2 polarized macrophages. The purpose of this microarray is to identify genes that may be differentially expressed in M1 or M2 macrophages after treatment with CNI-1493. CNI-1493 is a known inhibitor of M1 macrophages but details of its molecular mechanism are unknown. The effect of CNI-1493 on M2 macrophages has yet to be explored, but we hypothesize that CNI-1493 treatment will attenuate pro-tumor properties of M2 macrophages. We demonstrate with this array that known macrophage markers are unchanged after treatment with CNI-1493, indicating that CNI-1493 does not change the macrophage phenotype on a transcriptional level. Additionally, no candidate genes to suggest how CNI-1493 may attenuate the pro-tumor effects of M2 macrophages are readily identifiable. Total RNA extracted from M1 or M2 macrophages after polarization with GM-CSF (25ng/ml) or M-CSF (25ng/ml) for 7 days, followed by addition of IFN-γ (20ng/ml) and LPS (100ng/ml) or IL-4 (40ng/ml) for 18 hours, respectively, from CD14+ human PBMCs, and treated with CNI-1493 (200nM)

Yeast beta-glucan diets alter bone-marrow derived macrophage function

Project description:Fungal beta-glucans are major drivers of trained immunity which increases long-term protection against secondary infections. Heterogeneity in beta-glucan source, structure and solubility alters interaction with the phagocytic receptor Dectin-1 and could impact strategies to improve trained immunity in humans. Using a panel of diverse beta-glucans we describe the ability of a specific yeast-derived whole-glucan particle (WGP) to reprogramme metabolism and thereby drive trained immunity in human monocyte-derived macrophages in-vitro and mice bone-marrow in-vivo. Presentation of pure, non-soluble, non-aggregated WGPs led to the formation of the Dectin-1 phagocytic synapse with subsequent lysosomal mTOR activation, metabolic reprogramming and epigenetic rewiring. Intraperitoneal or oral administration of WGP drove bone-marrow myelopoiesis and improved mature macrophage responses, pointing to therapeutic and food-based strategies to drive immune training. Thus, the investment of a cell in a trained response relies on specific recognition of beta-glucans presented on intact microbial particles through stimulation of the Dectin-1 phagocytic response.

Differentially polarized human monocyte-derived macrophages (M0, M1, M2/IL10, M2/IL4, TAM-like)

Project description:In order to assess whether differences in the transcriptomic profile of human tumor-associated macrophages vs. alveolar macrophages from adjacent non-tumor-tissue (GSE162669) were caused by factors secreted by tumor cells, primary human monocyte-derived macrophages (MDMs) were polarized towards a TAM-like phenotype by cultivating them in tumor cell-conditioned medium. A genome-wide comparison by bulk RNA-Seq confirmed a high similarity of ex vivo TAMs and in vitro polarized TAM-like macrophages. Project description:Despite the remarkable success of immunotherapy in many types of cancer, pancreatic cancer has yet to benefit. Innate immune cells are critical players in antitumor immunosurveillance. Recent studies have revealed that innate immune populations may possess a form of memory, termed trained immunity through transcriptomic, epigenetic, and metabolic reprograming. 1Here, we show that intraperitoneal administration of yeast-derived particulate β-glucan results in 90% of that dose trafficking to the pancreas. This trafficking leads to a robust influx of newly characterized innate immune cells with an inflammatory monocyte/macrophage phenotype into the pancreas in a CCR2 dependent manner. These cells also exhibit a trained immunity phenotype upon exposure to tumor-derived factors and show enhanced phagocytosis and ROS-mediated cytotoxicity against pancreatic tumors. In orthotopic models of pancreatic cancer, mice trained with β-glucan show significantly reduced tumor burden and prolonged survival, which is further enhanced when combined with anti-PD-L1 immunotherapy. Cumulatively, these findings highlight a novel trafficking mechanism of yeast-derived particulate β-glucan that incites a phenotype of trained immunity specifically in the pancreas that can be utilized as an innovative strategy to treat pancreatic cancer.

Harnessing the Pre-metastatic Niche Macrophages through the Induction of Trained Immunity to control Metastasis.

Project description:We demonstrate that a natural compund B-Glucan (WGP) can induce training of macrophages by upregulating the Sphingolipid biosynthesis pathway and more specifically, the accumulation of S1P, resulting in mitochondrial fission in macrophages. These trained macrophages upon re-stimulation with tumor cells or tumor-derived factors showed increased responsiveness by increased pro-inflammatory cytokine production, increased phagocytosis and cytotoxicity. In vivo training of mice with WGP resulted in the training of Lung Interstitial macrophages and controlled the metastasis to the lungs.

Project description:We reported exosome-guided phenotype switches between M1- and M2-polarized BMDMs. M1- or M2-polarized BMDMs were successfully reprogrammed to M2- or M1-phenotype via the treatment of exosomes obtained from M2- or M1-polarized BMDMs. In this uploaded information, the exosomes from M1- and M2-polarized BMDMs were analyzed by high-throughput sequencing.
